Engineering the anthocyanin regulatory complex of strawberry (Fragaria vesca)

Abstract: The woodland strawberry, Fragaria vesca is a model fruit for a number of rosaceous crops. We have engineered altered concentrations of anthocyanin in F. vesca, to determine the impact on plant growth and fruit quality. Anthocyanin concentrations were significantly increased by over-expression or decreased by knock-down of the R2R3 MYB activator, MYB10. In contrast, a potential bHLH partner for MYB10 (bHLH33) did not affect the anthocyanin pathway when knocked down using RNAi constructs. Metabolic analysis of f… Show more

“…A similar result also was observed for strawberry FaMYB1 . In fruit, FaMYB1 showed the highest transcription level at the red ripe stage when anthocyanin accumulation reached a peak (Aharoni et al ., ; Lin‐Wang et al ., ). These results suggest that the PpMYB18‐ like genes are likely to be induced by anthocyanin‐ and/or PA‐related activators, thus providing negative feedback regulation to MBW complexes.…”

“…18,19,21 In the prior studies, expression of multiple early- and late-regulated biosynthesis genes in the flavonoid/phenylpropanoid pathway was reduced in transgenic MYB10 RNAi or yellow-fruited strawberry. In contrast, our RNA-seq data showed a limited number of biosynthesis genes whose expression was reduced in the yellow-fruited strawberry.…”

“…Peach and octoploid strawberry ( Fragaria × ananassa ) genome encodes two and four copies of Ans gene, respectively (Almeida et al, 2007; Ravaglia et al, 2013). Only one gene copy has been described in diploid strawberry ( F. vesca ) and apple (Takos et al, 2006; Lin-Wang et al, 2014). …”
