Endosomal Deubiquitinating Enzymes Control Ubiquitination and Down-regulation of Protease-activated Receptor 2

Abstract: The E3 ubiquitin ligase c-Cbl ubiquitinates the G proteincoupled receptor protease-activated receptor 2 (PAR 2 ), which is required for postendocytic sorting of activated receptors to lysosomes, where degradation terminates signaling. The mechanisms of PAR 2 deubiquitination and its importance in trafficking and signaling of endocytosed PAR 2 are unknown. We report that receptor deubiquitination occurs between early endosomes and lysosomes and involves the endosomal deubiquitinating proteases AMSH and UBPY. Ex… Show more

“…5G). Our findings are in agreement with previous reports showing that depletion of USP8 or overexpression of a dominant negative USP8 mutant causes the accumulation of ubiquitinated proteins within endosomes and a corresponding increase in endosome size (24,31,34,36,41,42).…”

“…These abnormalities have been attributed to numerous causes. First, it has been attributed to the accumulation of ubiquitinated proteins within endosomes (24,34,36,41). Second, it has been attributed to the dysfunction of the endosomes due to loss of endosomal proteins like STAM and HRS (23,36).…”

“…Endosome-Along with published evidence (19,22,23,37), the observations described above suggest that USP8 may function as a general regulator of Hrs-dependent trafficking at the sorting endosome. To investigate the cellular interactions between USP8 and the ESCRT-0 complex, BiFC microscopy was employed.…”

“…The deubiquitinating enzyme, known as associated molecule with the SH3 domain of STAM (AMSH), has been implicated in the regulation of epidermal growth factor receptor, calcium-sensing receptor, and most recently, the ␦-opioid receptor and the protease-activated receptor 2 (37)(38)(39)(40)(41)(42)(43). AMSH was first identified as an interacting partner of the SH3 domain of STAM (44) and later characterized as a Zn 2ϩ -dependent ubiquitin isopeptidase with a substrate preference for Lys-63-linked polyubiquitin chains (37,45).…”
