Abstract: Glial cell line-derived neurotrophic factor (GDNF) activity in ventral tegmental area (VTA) mediates the time-dependent increases in cue-induced cocaine-seeking after withdrawal (incubation of cocaine craving). Here, we studied the generality of these findings to incubation of heroin craving. Rats were trained to self-administer heroin for 10 days (6-h/day; 0.075 mg/kg/ infusion; infusions were paired with a tone-light cue) and tested for cue-induced heroin-seeking in extinction tests after 1, 11 or 30 withdra… Show more

“…In studies on incubation of cocaine or heroin craving administration of BDNF or GDNF in the mesocorticolimbic system have been shown to increase craving (Airavaara et al, 2011a; Lu et al, 2004a; Lu et al, 2009). On the other hand, several alcohol studies have shown that GDNF has acute inhibitory effects on drug-seeking behavior (Carnicella et al, 2008; He and Ron, 2006).…”

“…In one study heterozygous GDNF+/− mice displayed enhanced conditioned place preference (Niwa et al, 2007) while others have reported the conditioned place preference to be similar in GDNF+/− and wild-type mice, but of shorter duration in GDNF+/− mice (Airavaara et al, 2007). In studies of heroin craving GDNF injections in the NAc, but not VTA, increase the extinction response after withdrawal (Airavaara et al, 2011a). Similar to the study by Li et al (2014) GDNF has been also earlier found to block morphine-induced tyrosine hydroxylase upregulation (Messer et al, 2000).…”

“…As technology has improved, neurosurgical interventions like deep brain stimulation (DBS) have had clinical application and have become United States Food and Drug Administration (US FDA) approved standard therapy in neuropsychiatric disorders like medication refractory clinical depression, Tourette syndrome, obsessive compulsive disorder, dystonia, tremor and PD as the risk versus benefit ratio for invasive therapy has improved dramatically (Altinay et al, 2015; Guercio et al, 2015; Hamani et al, 2014; Kohl et al, 2014; Morishita et al, 2014). Neurosurgical interventions like DBS and GDNF replenishment into the NAc have been tried successfully in rat models of cocaine addiction (Airavaara et al, 2011; Girard et al, 2006; Green-Sadan et al, 2003; Green-Sadan et al, 2005; Lu et al, 2009; Pickens et al, 2011; Vassoler et al, 2013). Moreover, there is growing consensus that the neurobiological basis for drug addiction is becoming more clear and interventions that correct this pathophysiology may have tangible clinical benefits (Benarroch, 2015; Guercio et al, 2015; Pereira et al, 2015; Yadid et al, 2013).…”

“…Overexpression or intra-VTA infusion of GDNF during withdrawal from cocaine self-administration increases cue-induced cocaine seeking, an effect that can be reversed by blocking VTA ERK signaling (Lu et al, 2009). Heroin self-administration and withdrawal regulate GDNF mRNA expression in the NAc and VTA in a time-dependent manner, whereas infusion of GDNF to the NAc, but not to the VTA, potentiates cue-induced heroin seeking (Airavaara et al, 2011). Despite this, infusion of anti-GDNF into the NAc or VTA had no effect on incubation of heroin craving.…”
