Elucidation of radiation-resistant clones by a serial study of intratumor heterogeneity before and after stereotactic radiotherapy in lung cancer

Abstract: Stereotactic radiotherapy (SRT) for inoperable stage I non-small cell lung cancer has shown promising results and is now an alternative therapy for this disease. Several reports have detailed changes in mutation profiles after treatment with chemotherapy; however, such changes after SRT for lung cancer have not been reported. A patient who received SRT for lung cancer developed local recurrence 9 months after treatment and underwent surgery in our department. Using bronchoscopically biopsied and surgically res… Show more

“…Toward this objective, examination of many surgical samples collected from a large patient cohort with similar clinical characteristics was required for effective analysis of prognosis. As gene sequencing on surgical samples is being aggressively performed at our hospital since July 2014, we had access to sufficient number of sequenced samples [ 12 , 13 , 14 , 15 , 16 , 17 , 18 , 19 ]. In this context, association between clinical and genomic characteristics was analyzed in this study.…”

“…This method accounts for allelic imbalances introduced by changes in the segment copy number and sample contamination by normal cells [ 40 ]. In this study, PyClone analysis was performed to estimate the fraction of cancer cells harboring mutant GTF2I [ 40 , 41 , 42 ].…”

“…Hierarchical clustering generates a binary tree, in which the most similar patterns are clustered. To organize tumor mutational profiles into meaningful structures based on similarity or dissimilarity, we used unsupervised hierarchical clustering with average and complete linkage algorithms in GeneCluster, applying the same approach as previously reported [60,61,62,63]. This process places cases with similar mutation profiles as neighboring rows in the clustergram.…”
