Due to their adjuvant effect and their ability to chaperone tumor-associated peptides, heat shock proteins constitute a potent alarm signal for the immune system and can lead to activation of anti-tumor T-cell immunity.Radiofrequency ablation has been reported to induce heat shock protein expression especially that of heat shock protein 70 in sublethally damaged tumor cells. Heat shock protein 70 (Hsp70) release and its effects on pro-inflammatory cytokine production have been controversial. In this study, we investigated whether Hsp70 could be released from monocytes and activates matrix metalloproteinase-9 (MMP-9) gene expression. Hsp70 overexpression in human monocytic cell line U937 was found to increase PMAinduced MMP-9 expression and enhance cell motility. Hsp70 cDNA transfectants released Hsp70 protein into culture supernatants, and a part of released Hsp70 subsequently was bound to the surface of U937 cells. Addition of culture medium containing the extracelluar Hsp70 led to an increase not only in proMMP-9 secretion, but also the invasiveness of U937 cells through Matrigel or human umbilical vascular endothelial cells (HUVEC) in vitro. Immunodepletion of Hsp70 abolished its effect on MMP-9 expression. The released Hsp70 activated nuclear factor kappa B (NF-κB) and activating protein-1 (AP-1), which led to the activation of MMP-9 transcription. Taken together, these results suggest that extracellular Hsp70 induces the expression of MMP-9 gene through activation of NF-κB and AP-1.

ÖZAmaç: Hafif preeklamptik hastalar, şiddetli preeklamptik hastalar ve normotansif gebelerden seçilmiş kontrol grubunda, serum ısı şok protein 70 (HSP70) konsantrasyonlarının farklı olup olmadığının araştırılması. Yöntem: Çalışmamızda Atatürk Üniversitesi Tıp Fakültesi Kadın Hastalıkları ve Doğum Kliniğine başvuran 18-45 yaş arası, 20. gestasyonel haftanın üzerindeki monofetüs gebeliğe sahip 30 hafif preeklamptik, 30 şiddetli preeklamptik hasta ile kontrol amaçlı seçilen poliklinkte takip edilmekte olan 30 normotansif sağlıklı gebe, randomize prospektif olarak incelendi. Serum HSP70 konsantrasyonları ELISA yöntemi ile ölçüldü. Bulgular: Çalışma sonuçları incelendiğinde, hafif preeklamptik hastaların HSP70 değerle-ri: 0,634 ng/mL±0,30, şiddetli preeklamptik hastalarda 0,881 ng/mL±0,37, kontrol grubunda ise 0,619 ng/mL±0,27 olarak saptandı. Şiddetli preeklamptik grubun değerlerinde hafif preeklamptik gruba göre anlamlı yükseklik mevcuttu (p=0,01, GA: 0,0467-0,4483), şiddet-li preeklamptiklerin kontrol grubu ile kıyaslanmasında da yine yüksek değerler tespit edildi (p=0.006, GA: 0,0612-0,4628). Korelasyon analizlerinde ise HSP70 düzeyleri ile sistolik kan basıncı (r=0,325, p=0,002); diastolik kan basıncı (r=0,273, p=0.009) ve 24 saatlik idrarda protein (r=0,291, p=0,005) düzeyleri ile anlamlı pozitif korelasyonlar tespit edildi. Sonuç: Çalışmamız sonucunda şiddetli preeklamptik hastaların serum HSP70 düzeyleri hafif preeklamptik hastalara göre ve kontrol grubuna göre daha yüksek saptandı. Preeklampsi ve komplikasyonlarını öngörme konusunda HSP70 önemli bir gösterge olabilir, ancak bu konuda daha kapsamlı çalışmalara gereksinim vardır.Anahtar kelimeler: Gebelik, preeklampsi, ısı şok protein70, HSP 70, HSPA ABSTRACT Objective: The aim of this study was to evaluate whether serum levels of heat shock protein 70 (HSP70) in patients with mild preeclampsia, severe preeclampsia differ when compared with normotensive pregnant controls. Methods: In this randomize prospective study, we enrolled a total of 90 monoparous (30 mild, and 30 severe preeclamptic patients, and 30 normotensive healthy pregnant controls) pregnants aged 18-45 years who passed their 20. gestational weeks, and admitted to the Clinics of Obstetrics and Gynecology of Atatürk University Hospital. Serum HSP70 concentrations were measured by standard ELISA techniques. Results:The mean values of serum HSP70 in mild (0.634±0.30 ng/ml), and severe (0.881±0.37 ng/ml) preeclamptic patients and normotensive controls (0.619±0.27 ng/ml) were measured. Severe preeclamptic patients had statistically significantly higher concentrations of serum HSP70 when compared with mild preeclamptic patients (p=0.01; CI: 0.0467-0.4483), and controls (p=0.006; CI: 0.0612-0.4628). We found that HSP70 concentrations positively correlated with systolic (r=0.325; p=0.002), and diastolic blood pressures (r=0.273; p=0.009) and daily urinary protein (r=0.291; p=0.005) levels.
