Electrografted Diazonium Salt Layers for Antifouling on the Surface of Surface Plasmon Resonance Biosensors

Abstract: Electrografted diazonium salt layers on the surface of surface plasmon resonance (SPR) sensors present potential for a significant improvement in antifouling coatings. A pulsed potential deposition profile was used in order to circumvent mass-transport limitations for layer deposition rate. The influence of number of pulses with respect to antifouling efficacy was evaluated by nonspecific adsorption surface coverage of crude bovine serum proteins. Instead of using empirical and rough estimated values, the pene… Show more

“…3B). 57 Surface coverage due to fouling by crude bovine serum (undiluted, 75 mg protein per mL), measured by SPR upon 20 min exposure, was relatively good at Γ = 42 ± 4 ng cm −2 . 54,55 For the nine different peptide designs investigated, NSA was consistently lower than Γ = 80 ng cm −2 , highest resistance to serum adsorption being the feat of 3-MPA-Ser 3 -Asp 2 -OH SAMs with Γ = 23 ± 10 ng cm −2 (Fig.…”

“…The following year, the investigators were able to substantially decrease NSA by expanding the scope of their study to short 3-MPA-homopeptide-based SAMs. 57 In this 2015 account, anchorage to gold substrates did not rely on thiol chemistry however, 48,52 but on an electrografting reaction involving a diazonium salt of phenylalanine (Fig. 3B).…”

“…Other significant examples are the construction of SPR platforms based on aryl dizonium electrographting to achieve a significant improvement in antifouling coatings [71]. Pairwise antibody footprinting is also a relevant approach for the direct immobilization of antibodies to block the unoccupied immobilized anti-Fc to prevent non-specific antibody binding [72].…”

“…Label-free detection platforms, which include surface plasmon resonance (SPR) (Zou et al, 2015), quartz crystal microbalance (QCM) (Tang et al, 2013), atomic force microscopy (AFM) (Zhang et al, 2014), and electrochemical impedance spectroscopy (EIS) (Date et al, 2014), show tremendous promise for quantitative protein detection. Label-free processes are usually simple, cost-effective, less time-consuming, and amenable to real-time analysis, thereby improving assay efficiency (Okuno et al, 2007; Zhao et al, 2009a; Qi and Li, 2011; Pang et al, 2015).…”
