Introduction: Development of a new enzymatic kinetic-photometric method for analysis of Dequalinium chloride, based on the use of enzymatic hydrolysis of acetylcholine, for determine the amount of Dequalinium chloride in the vaginal tablets “Fluomizin” No 6. Method: The amount of Dequalinium chloride (DCh) was determined by the degree of inhibition of the enzymatic reaction, which was evaluated by the residual unreacted substrate - acetylcholine. Determination of the residual amount of acetylcholine in the reaction mixture was performed by a kinetic-photometric method using an indicator oxidation reaction of p-phenetidine with peracetic acid, which is formed during the auxiliary reaction of perhydrolysis with addition of excess hydrogen peroxide in the reaction mixture over a period of time. Results: Optimal conditions were determined: pH 8.35 was selected, influence of nature of buffer solution, concentration of acetylcholine (0.05 mg / ml), cholinesterase (0.4 mg / ml), hydrogen peroxide (10,0%) and p-phenethedine (1 %), the incubation time (10 min). Conclusions: The procedure for determination of Dequalinium chloride content in the vaginal tablets “Fluomizin” No 6 was developed. The kinetic parameters Km (Michaelis–Menten constant) and Vmax (maximal velocity) were determined.

Розробка та валідація ензимної кінетико-фотометричної методики визначення залишкових кількостей деквалінію хлориду на поверхні фармацевтичного обладнання Мета. Розробка та валідація нової ензимної кінетико-фотометричної методики аналізу деквалінію хлориду, що ґрунтується на використанні реакції ферментного гідролізу ацетилхоліну, для визначення залишкових кількостей деквалінію хлориду при здійсненні контролю повноти очищення фармацевтичного обладнання. Результати та обговорення. Встановлені оптимальні умови перебігу ензимної реакції-порядок змішування та концентрації ацетилхоліну (0,05 мг/мл), холінестерази (0,4 мг/мл), водню пероксиду (10 %) та п-фенетидину (1 %), час витримування реакційної суміші (20 хв), рН середовища (8,35), вплив природи буферного розчину. Проведено валідацію розробленої методики-запропоновано діапазон застосування методики (40-160 % в нормалізованих координатах, за 100 % обрано максимально допустиму концентрацію деквалінію хлориду в змивах з фармацевтичного обладнання х crit = 0,5 мкг/мл), кількість концентраційних рівнів (n = 7) та порядок їх розташування всередині діапазону застосування, з урахуванням чого розраховано максимально допустиму невизначеність методики (maxΔ x = 3,05 %) і, відповідно, критерії прийнятності параметрів лінійної залежності, систематичної та випадкової похибок. Показано, що валідаційні характеристики розробленої методики відповідають розрахованим критеріям прийнятності. Встановлено ступінь екстракції деквалінію хлориду зі свабу зі змивом з фармацевтичного обладнання. Експериментальна частина. Визначення залишкових кількостей деквалінію хлориду на поверхні фармацевтичного обладнання визначали за ступенем інгібування ензимної реакції, який оцінювали за залишком субстрату біохімічної реакції-ацетилхоліну, що не прореагував. Визначення залишкової кількості ацетилхоліну в реакційній суміші виконували кінетико-фотометричним методом за індикаторною реакцією окиснення n-фенетидину надацетатною кислотою (утворюється під час допоміжної реакції пергідролізу при додаванні до реакційної суміші надлишку водню пероксиду), реєструючи світлопоглинання утворюваного продукту реакції-азоксифенетолу (λ max = 358 нм) впродовж певного періоду часу. Висновки. Розроблено ензимну кінетико-фотометричну методику визначення залишкових кількостей деквалінію хлориду на поверхні фармацевтичного обладнання після його очищення. Проведено валідацію розробленої методики за такими параметрами, як лінійність, правильність, прецизійність та межа кількісного визначення.

Goal. Optimization of the effectiveness of diagnosis and prediction of the degree of risk of violation of the vaginal microbiota.Materials and methods. 572 patients were examined with complaints of copious discharge from the genital tract and/or an unpleasant smell. All women were observed in accordance with anamnestic, clinical, and laboratory criteria.Results and conclusions. When examining patients, the results of anamnestic, clinical and laboratory signs were evaluated according to a point system, taking into account their informative value and the probability of influencing the vaginal microbiome. Based on the data analysis, a method for predicting the degree of risk of violation of the vaginal microbiota was proposed. The method allows for a personalized therapeutic and diagnostic approach and allows predicting the risk of violation of the vaginal microbiota as early as possible.
