Orthodontics is constantly seeking innovation towards mechanical efficiency and better oral-related quality of life during treatment. This narrative review aims to discuss novel scientific reports about light therapies and how they can optimize different stages of orthodontic intervention: before, during, and after treatment. Recurrent conditions that can be treated with laser devices are the removal of carious tissue, dentin hypersensitivity, and temporomandibular disorders. Evidence reveals that laser procedures accelerate health recovery, enabling individuals to initiate orthodontic treatment. Along orthodontic procedure, photobiomodulation therapy, is indicated for analgesia after appliance activations, repair of traumatic ulcers, and acceleration of tooth movement. Moreover, antimicrobial photodynamic therapy is well-indicated for effective decontamination of oral infections such as herpetic lesions and peri-implantitis. Finally, high-intensity lasers are good allies in removing brackets and reconditioning red esthetics. There are many benefits to the use of light sources in the orthodontic routine: simplicity of technique, ease of handling devices, minimal invasiveness, and patient comfort during procedures. It is essential that professionals develop a critical overview of technological advances, offering safe and evidence-based therapies. Recent advances indicate that laser therapies improve patient experiences during orthodontic treatment and minimize the side effects of clinical interventions.

A set of arylazo sulfones, known to undergo N–S bond cleavage upon light exposure, has been synthesized, and their activity in the dark and upon irradiation towards DNA has been investigated. Their interaction with calf-thymus DNA has been examined, and the significant affinity observed (most probably due to DNA intercalation) was analyzed by means of molecular docking “in silico” calculations that pointed out polar contacts, mainly via the sulfonyl moiety. Incubation with plasmid pBluescript KS II revealed DNA cleavage that has been studied over time and concentration. UV-A irradiation considerably improved DNA damage for most of the compounds, whereas under visible light the effect was slightly lower. Moving to in vitro experiments, irradiation was found to slightly enhance the death of the cells in the majority of the compounds. Naphthylazosulfone 1 showed photo-disruptive effect under UV-A irradiation (IC50 ~13 μΜ) followed by derivatives 14 and 17 (IC50 ~100 μΜ). Those compounds were irradiated in the presence of two non-cancer cell lines and were found equally toxic only upon irradiation and not in the dark. The temporal and spatial control of light, therefore, might provide a chance for these novel scaffolds to be useful for the development of phototoxic pharmaceuticals.

Background: The complex traditional treatment of inflammation diseases in oral cavity includes the prescription of antibiotic and antiseptic therapy. This systematic review aims to evaluate the effect of photodynamic therapy as a part of management of inflammatory diseases in oral cavity; Methods: The study is presented in accordance with the preferred reporting points for systematic reviews and meta-analyses (PRISMA). This systematic review was conducted using electronic databases such as Medline PubMed, Scopus and the Cochrane Central Register of Controlled Trials. All the studies in this systematic review, were randomized, the risk of bias 2 (ROB 2) were assessed; Results: Considering the inclusion and exclusion criteria, we included 10 randomized clinical trials, published up to 2023 investigating the application of photodynamic therapy as a part of management of inflammatory diseases in oral cavity. The diode laser was used in the oral cavity in the zone of inflammatory process (gingivitis, mucositis, periimplantitis, marginal periodontitis, abscess, periostitis, osteomyelitis etc.) in nine studies or in the zone before surgical procedures in one study; Conclusion: Based on the results of clinical studies, it can be stated that photodynamic therapy shows good results for operations performed in the oral cavity and pharynx.
