Efficacy of a novel topical fipronil, (S)-methoprene, eprinomectin and praziquantel combination against naturally acquired intestinal nematode and cestode infections in cats

Abstract: The efficacy of a novel topical combination formulation of fipronil, (S)-methoprene, eprinomectin and praziquantel against naturally acquired intestinal nematode and cestode infections in cats was evaluated in seven negative control, blinded studies. Cats were selected based on a pre-treatment faecal examination indicating a patent infection with at least hookworms (two studies), Toxocara ascarids (one study), taeniid cestodes (two studies) or Dipylidium cestodes (two studies). In each study, cats were assigne… Show more

“…Eprinomectin and praziquantel plasma profiles following a topical application of the combined formulation have systemic endoparasiticide concentrations needed to kill nematodes and cestodes. Both compounds also have appropriate PK parameters (AUC, C max and T max ), that are identical to those of Broadline Ò , a product with proven efficacy against nematodes and cestodes [12,29,35]. This was confirmed by efficacy data obtained with NexGard Ò Combo against both nematodes and cestodes [13] in cats.…”

“…Eprinomectin is an avermectin, from the macrocyclic lactone class, binding selectively to glutamate-gated chloride ion channels on nerves and muscular cells of several types of invertebrates, including nematodes. Eprinomectin is a well-known compound used in cattle, sheep and goats [27,33], and cats [12,29]. Praziquantel is a pyrazino-isoquinoline derivative anthelminthic that acts specifically on cestodes and trematodes, and is a well-known compound in veterinary and human medicine, in oral and topical forms [2,3,13,29,32,35,39].…”

“…Fipronil/ (S)-methoprene and esafoxolaner provide insecticide and acaricide properties, eprinomectin and praziquantel provide nematodicide and cestodicide properties, respectively. Broadline Ò has been used safely for several years [8] and was demonstrated to be efficacious against a broad spectrum of feline parasites, including fleas [1] and gastro-intestinal helminths [13,24]. NexGard Ò Combo was recently developed and registered in the European Union [9] and was demonstrated to be efficacious against fleas in experimental infestation studies conducted with isolates originating from France, South Africa and the USA [25], and in field studies with naturally infested cats from Europe, the USA and Australia [27], and against gastrointestinal helminths using induced and natural models of infection with isolates originating from Europe, North America and South Africa [12].…”
