Abstract: Background Knee osteoarthritis (OA) is one of the most common joint diseases, and pharmacotherapy is necessary to control its symptoms. Aim To evaluate efficacy and tolerability of duloxetine in patients with knee OA. Methods PubMed, Web of Science, Embase, Cochrane Library and http://ClinicalTrials.gov were searched to identify randomised controlled trials comparing duloxetine with placebo for knee OA. Data including pain, stiffness, physical function, and adverse events were extracted for meta‐analysis. The … Show more

“…Improvement in physical function is also important in patients with OA or CLBP. Our results showed that duloxetine had significant improvement over placebo in both the WOMAC physical function subscale and RMDQ-24, which is consistent with previous metaanalyses that evaluated duloxetine in the treatment of OA 23,26,27 .…”

“…In addition, the guidelines recommend against or cautious use of antidepressants for CLBP 8 . Recently, two meta-analyses of duloxetine in the treatment of OA were published almost simultaneously 26,27 . However, both of the studies focused on knee OA limited by the number of articles included.…”

“…Among available and established pharmacological treat ments, antidepressants are effective in different chronic musculoskeletal pain conditions associated with central sensitisation, such as fibromyalgia, 81 low back pain, 82 neck pain, 83 and knee osteoarthritis. 84 A syste matic review found moderate to high quality evidence that the anti convulsant gabapentin is ineffective for the treatment of low back pain or lumbar radicular pain, 85 whereas another systematic review found insuffi cient evidence to either support or refute the efficacy of gabapentin in reducing pain in patients with fibromyalgia. 86 Three randomised controlled trials found pregabalin to be effective in reducing pain associated with fibromyalgia.…”

“… 26 The third system review and meta-analysis published in 2019 is effective in the management of chronic pain and loss of physical function but having no advantage in treating joint stiffness. 27 The consistent findings from above system reviews and meta-analyses were duloxetine had a beneficial impact on pain relief, function improvement across all studies. In addition, our evidence showed that duloxetine can also ameliorate the knee stiffness, although the significant change in the quality of life cannot be verified.…”
