Poor germination percentage and non-uniform seedling emergence and establishment are major causes of low rice yield in water-deficit arid ecosystems. A seed priming study with a low concentration of MoP, DAP, urea, vitamin C, ZnSO4, sucrose, NaCl, and hydro priming was conducted to assess the seed germination, seedling emergence, establishment, and seedling growth of the spring rice variety Hardinath-1. The experiment was conducted using a completely randomized design with three replications. Two test sets of experiments were carried out using blotting paper rolls and separate sand trays. Germination was observed at intervals of 24 hours up to 7 days, and germination and growth parameters were monitored. The results showed that germination (88.67%) and seedling emergence (88.67%) were high in vitamin C at 7 days after sowing (DAS). Germination energy (81%), germination speed (91.78%), and vigor index (91.78) were higher with seed priming of MoP. The lowest germination (56.44%) was observed with the priming of urea. At 30 DAS, the longest shoot length (19.93 cm) and longest root length (11.79 cm) were observed with the priming of ZnSO4. The highest seedling fresh weight (0.75 g) and dry weight (0.16 g), the highest number of leaves (3.57), and the longest leaf length (11.97 cm) were found with the priming of MoP, NaCl, and ZnSO4, respectively. However, the lowest seedling growth parameters were observed in the control priming. The overall results revealed that seed priming with MoP is an effective tool to enhance the germination percentage and seedling establishment of rice. SAARC J. Agric., 21(2): 81-93 (2023)

The increasing world population needs a standard balanced diet to address malnutrition problems. For this purpose, seed priming is one of the best techniques, which helps to increase the production of functional and nutritional food crops. Different techniques have been used for seed priming, but biological priming is the most frequently used because biocontrol agents offer a friendly environment for the growth of food crops. In this study, Momordica charantia L. seeds were subjected to a strain of Enterobacter sp. FD17 as a biocontrol agent at different time exposures (i.e., 24 h, 48 h, and 72 h). Leaf growth, flavonoids, chlorophyll content, amino acids, soluble sugars, protein, and total soluble phenolics were studied in the vegetative stage. The yield of nutritive components was evaluated from fruit, peel, and pulp of M. charantia. Biopriming was revealed to improve the final emergence rate, mean emergence time, seedling vigor, emergence index, and vigor indices I and II. Among the growth parameters, the root (0.45 ± 0.045 g) and shoot fresh weight (1.23 ± 0.05 g), leaf area (15.52 ± 1.5 cm), shoot length (30.33 ± 0.58 cm), number of flowers (6 ± 1.0), fruit weight (96.33 ± 1.15 g), and germination percentage (56.67 ± 11.55%) were also improved. Among biochemical analyses, biopriming improved chlorophyll a (6.33 ± 0.58 mg/g) and b (8.58 ± 2.5 mg/g), total soluble sugar (33.13 ± 2.24%), and total chlorophyll content (9.0 ± 1.5 mg/g). The nutritional analysis showed that free amino acids (1.43 ± 0.02 mg/g), total soluble sugar (42.53 ± 1.65%), ash (20.53 ± 2.57%), and catalase (347.47 ± 34.76 U/g) were increased in fruit, while crude fiber (3.62 ± 0.1%) and peroxidase (5.61 ± 0.34 U/g) in peel and protein and metabolizable energy in peel and fruit were increased. Among the water, acetone, and methanol extracts, the maximum antibacterial activity was shown by methanol extracts of leaves against Gram-negative and Gram-positive bacterial species (i.e., Pseudomonas aeruginosa and Staphylococcus aureus, respectively) with inhibitory diameters of 3 mm. Biopriming also improved the phenolic contents in the leaves and fruits of M. charantia. Biopriming treatment was also revealed to be directly correlated with antiglycation activity. Therefore, biopriming treatment on seeds could be used to manipulate plant cell metabolism with a substantial improvement in phenolic content, antibacterial activity, and growth of M. charantia.

Endophytic Fusarium oxysporum GW controlling weed and an effective biostimulant for wheat growth

Wheat crop has to compete with several weeds including Avena fatua, a noxious weed that alone is responsible for 30–70% losses in the yield annually. Because of the environmental concerns associated with conventional methods, researchers are on a continuous hunt to find clean alternatives in order to manage weeds. Fungi have shown promising weedicide potential in lab studies. The current study aimed to isolate endophytic fungi from wheat plants which can promote wheat growth and inhibit the growth of common weed, A. fatua. Of several isolates, GW (grayish white) was selected for its promising features, and the strain was identified as Fusarium oxisporum through ITS sequencing technique. This fungus released a number of compounds including Isovitexin, Calycosin, quercetagetin, and dihydroxy-dimethoxyisoflavone that inhibited the growth of A. fatua but did not influence the growth of wheat seedlings. Biomass of this fungus in the soil also reduced growth parameters of the weed and promoted the growth of wheat. For instance, the vigor index of A. fatua seedlings was reduced to only 6% of the control by this endophyte. In contrast, endophyte-associated wheat seedlings showed a higher vigor index than the control. Behind this differential response of the two plants were their contrasting physiological and biochemical status. Lower growth phenotypes of A. fatua seedlings had reduced levels of IAA, GAs, and SA and higher the levels of JA and ABA. Besides, their ROS scavenging ability was also compromised as evident from relatively lower activities of catalase, peroxidase, and ascorbic acid oxidase, as well as higher accumulation of ROS in their leaves. Wheat seedlings response to GW was opposite to the A. fatua. It may be concluded that F. oxysporum GW has the ability to differentially modulate physiology and biochemistry of the two hosts leading to contrasting phenotypic responses.
