Respiratory infection can be exacerbated by the high glucose concentration in the airway surface liquid (ASL). We investigated the effects of salbutamol and phlorizin on the pulmonary function, oxidative stress levels and SGLT1 activity in lung, pulmonary histopathological damages and survival rates of rats with sepsis. Sepsis was induced by cecal ligation and puncture surgery (CLP). Twenty-four hours after surgery, CLP rats were intranasally treated with saline, salbutamol or phlorizin. After 2 hours, animals were anesthetized and sacrificed. Sepsis promoted atelectasis and bronchial inflammation, and led to increased expression of SGLT1 on cytoplasm of pneumocytes. Salbutamol treatment reduced bronchial inflammation and promoted hyperinsuflation in CLP rats. The interferon-ɤ and Interleucin-1β concentrations in bronchoalveolar lavage (BAL) were closely related to the bronchial inflammation regulation. Salbutamol stimulated SGLT1 in plasma membrane; whereas, phlorizin promoted the increase of SGLT1 in cytoplasm. Phlorizin reduced catalase activity and induced a significant decrease in the survival rate of CLP rats. Taken together, sepsis promoted atelectasis and lung inflammation, which can be associated with SGLT1 inhibition. The loss of function of SGLT1 by phlorizin are related to the augmented disease severity, increased atelectasis, bronchial inflammation and a significant reduction of survival rate of CLP rats. Alternatively salbutamol reduced BAL inflammatory cytokines, bronchial inflammation, atelectasis, and airway damage in sepsis. These data suggest that this selective β2-adrenergic agonist may protect lung of septic acute effects.

In British Columbia (BC), harmful algal blooms (HABs) regularly cause severe economic losses through finfish mortalities and shellfish harvest closures due to toxin accumulation, gill damage, or hypoxia. As there is no routine governmental monitoring of HAB phenomena in BC, HAB variability, and its potential links to environmental drivers are not well understood. Here we present results from a well-managed citizen science program which collected an unprecedented 4 year, high-resolution (∼bi-monthly, ∼80 stations) dataset of harmful algae (HA) concentrations and corresponding physical and chemical properties of seawater throughout the Strait of Georgia (SoG), BC. Analysis of this dataset revealed statistically significant interannual and seasonal relationships between environmental drivers and the most common HA taxa: Rhizosolenia setigera, Dictyocha spp., Alexandrium spp., Heterosigma akashiwo, Chaetoceros convolutus, and C. concavicornis. HABs exhibited significant interannual variations; specifically, no HABs were found during the summer of 2015, blooms of Dictyocha occurred in 2016 and 2017, and dense blooms of Heterosigma and Noctiluca occurred in 2018. In addition, HA prevalence corresponded with negative effects observed in local aquaculture facilities where higher toxins concentrations (causing Paralytic and Diarrhetic Shellfish Poisonings) in shellfish flesh were detected during years with greater abundance of Alexandrium and Dinophysis. Furthermore, salmon mass mortality at fish farms corresponded to years with high concentrations of Heterosigma and Dictyocha. As such, these results highlight the need for long-term data to evaluate the potential role of HA as a stressor on the SoG ecosystem.

Atherosclerosis is a multifactorial disorder, which affects the arterial wall. It has been reported that, hypothyroidism and thyroid hormone deficiency are related to cardiovascular disorders. Also, endothelial dysfunction plays an essential role in the development of atherosclerosis. We aimed to evaluate the effects of thyroid-stimulating hormone (TSH) on pro-inflammatory tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), angiogenic vascular endothelial growth factor (VEGF) and leukocyte adhesion, intercellular adhesion molecule 1 (ICAM-1) and E-selectin in human umbilical vein endothelial cells (HUVECs). In this study, HUVEC cells were treated with 1 and 2 μM of TSH in different treatment times. The gene and protein expression of ICAM-1, VEGF, and E-selectin were performed by real-time polymerase chain reaction and western blotting, respectively. Likewise, TNF-α and IL-6 protein levels were determined by the ELISA method. VEGF, ICAM-1, and E-selectin as endothelial dysfunction markers and also, TNF-α and IL-6 as pro-inflammatory cytokines were detectable in HUVEC. Besides, the results of this study revealed that TSH treatment down-regulates TNF-α and IL-6. Evaluating the gene and protein expression data revealed the upregulation of ICAM-1, E-selectin, and VEGF in TSH treated cases in different periods of exposure. Considering the multiple actions of TSH, it could be concluded that TSH plays a controversial role in atherogenesis by anti-inflammatory effects and on the other side, angiogenesis and leukocyte adhesion induction which is related to vascular cell proliferation.
