Effects of methomyl on lipid peroxidation and antioxidant enzymes in rat erythrocytes: In vitro studies

Abstract: Erythrocytes are a convenient model to understand the membrane oxidative damage induced by various xenobiotic pro-oxidants. This study was designed to investigate the possibility of methomyl (Lannate 90% SP), S-methyl N-(methylcarbamoyloxy) thioacetimidate, to induce oxidative stress response in rat erythrocytes in vitro. Erythrocytes were incubated for 4 hours at 37 degrees C with different concentrations (0.0, 0.1, 0.5, 1.0, 1.5 and 2.0 mM) of methomyl. The results showed that methomyl decreased acetylcholin… Show more

“…CPF-O can also be metabolized by esterases to form TCP and diethylphosphate (Chanda et al, 1997;Pond et al, 1998). Recent studies showed that CPF metabolism produced elevated ROS that generated oxidative stress in different cell types (Verma et al, 2007;Mansour et al, 2009;Saulsbury et al, 2009). In the present study, Fig.…”

“…More so, the erythrocyte membrane has been shown to be highly sensitive to oxidative stress as it contains high amount of polyunsaturated fatty acids as well as higher concentration of oxygen and heme [56]. On the other hand, erythrocyte osmotic fragility test gives an in-vitro measure of the tensile strength of erythrocyte membrane and it is an indirect method of evaluating lipid peroxidation in animals [57].…”

“…Highly reactive oxygen metabolites, especially hydroxyl radicals, act on unsaturated fatty acids of phospholipid components of membranes to produce malondialdehyde, a lipid peroxidation product. Previous studies indicate that insecticides in both in vivo [10,11,42] and in vitro tests [43] alter the enzyme activities associated with antioxidant defense mechanisms.…”
