Effects of extracellular calcium on cell membrane resealing in sonoporation

Abstract: Sonoporation has been exploited as a promising strategy for intracellular drug and gene delivery. The technique uses ultrasound to generate pores on the cell membrane to allow entry of extracellular agents into the cell. Resealing of these non-specific pores is a key factor determining both the uptake and post-ultrasound cell survival. This study examined the effects of extracellular Ca 2+ on membrane resealing in sonoporation, using Xenopus oocytes as a model system. The cells were exposed to tone burst ultra… Show more

“…At least in principle, this could allow also for separation of cells in which the membrane electrical properties are altered only temporarily, e.g., due to membrane poration by electric field [44], ultrasound [45], or chemical agents (e.g., actinoporins [46] or detergents [5]). The separation efficiency under given experimental conditions could also be improved by optimization of the electric field frequency used for dielectrophoresis under those particular conditions.…”

“…The fast resealing of the cell membrane poration was demonstrated by rapid decline of intracellular calcium levels [50,57] and restoration of cell membrane potential [55,[62][63][64].…”

“…We have previously employed a voltage clamp technique to monitor sonoporation (23)(24)(25)(26) in real time by measuring the transmembrane current (TMC) of single Xenopus oocytes. The ultrasound-induced localized membrane disruption allowed ions to flow nonselectively through the membrane, resulting in an increase of the TMC.…”
