Effects of cisplatin and thiosulfate upon auditory brainstem responses of guinea pigs

Abstract: Two side effects which limit the use of cisplatin in cancer chemotherapy are severe nephrotoxicity and ototoxicity. The concurrent administration of sodium thiosulfate with cisplatin reportedly protects from cisplatin nephrotoxicity, however, protection from ototoxicity has not been documented. The purpose of this study was to examine the efficacy of using thiosulfate to ameliorate the ototoxic effects of cisplatin. Toward this end, the effects of cisplatin alone, cisplatin administered concurrently with sodiu… Show more

“…For example, otoprotection has been observed for sodium thiosulfate (Church et al 1995;Otto et al 1988), diethyldithiocarbamate (Church et al 1995;Rybak et al 1995), glutathione (Campbell et al 1996(Campbell et al , 2003a, and D-methionine (Campbell et al 1999(Campbell et al , 2003b. To the best of our knowledge, there are only two previously published studies with proper scientific control groups that investigated WR's chemoprotective effects against CIS-induced ototoxicity (Church et al 1995;Kaltenbach et al 1997).…”

“…Sodium thiosul-fate protects against carboplatin ototoxicity in both humans and animals [Neuwelt et al, 1996[Neuwelt et al, , 1998Otto et al, 1988]. In addition, 4-methylthiobenzoic acid and diethylthiocarbamate not only protect against cisplatin ototoxicity in rats, but do so by maintaining the integrity of cellular antioxidant defense systems.…”

“…Sodium thiosulfate has been shown to protect against carboplatin ototoxicity in both animal and human studies [Neuwelt et al, 1996[Neuwelt et al, , 1998Otto et al, 1988]. Similarly, D-methionine (D-Met) has been shown to protect rats from the ototoxic effects of cisplatin [Campbell et al, 1996].…”

“…Sulfur containing compounds such as sodium thiosulfate (STS) and diethyl-dithiocarbamate (DDTC) were reported to provide good otoprotection against cisplatin (Otto et al, 1988), but had significant side effects and/or interference with antitumor activity (Jones et al, 1991;Church et al, 1995;Gandara et al, 1995;Muldoon et al, 2000). D-Methionine (D-Met), also a sulfur-containing nucleophilic antioxidant, has shown to provide effective nephroprotection against cisplatin without interfering with antitumor action (Jones and Basinger, 1989).…”
