Abstract: It is concluded that the HCO(3)(-) secretion stimulated by low concentrations of bile acids acts to protect the pancreas against toxic bile, whereas inhibition of HCO(3)(-) secretion by high concentrations of bile acids may contribute to the progression of acute pancreatitis.

“…Using the isoform-specific NHE inhibitor HOE-642 we also showed that the functions of NHE1-3 isoforms were reduced in response to 0.3mM CDC. Similarly, high concentration of CDC strongly inhibited the acid/base transporters of guinea pig pancreatic ductal epithelial cells [43].…”

“…Cl -withdrawal technique was used to investigate the activity of CBE [43]. Removing Cl -from the standard HCO 3 -/CO 2 buffered solution caused alkalization due to the reversed activity of the Cl -/HCO 3 -exchanger.…”

“…2) and largely inhibited by luminal 200 M H 2 DIDS. H 2 DIDS-sensitive apical Cl Ϫ / HCO 3 Ϫ exchange in guinea pig pancreatic duct was recently shown to be stimulated by bile acids by a mechanism dependent on intracellular Ca 2ϩ increase (41). mRNAs encoding both apical exchangers Slc26a3 and Slc26a6 have been detected in the mouse pancreatic duct and in the human pancreatic cell line CFPAC-1 (10,22,32).…”

“…Therefore, measuring the F340/F380 ratio is more sensitive than F340 alone, since not only the change in F340 corresponds to the intracellular ion concentrations but the inverse change in F380 as well, since the concentration of the dye changes too. One measurement was obtained per second (40). Compositions of solutions are detailed in Table 2.…”
