Abstract. The aim of the present study was to investigate the treatment effects and associated mechanism of Bak Foong pills (BFPs) combined with metformin in the treatment of polycystic ovarian syndrome (PCOS). BFPs and/or metformin were administrated to treat the PCOS rats, and the weights and morphologies of the ovary, uterus and adrenal gland were measured. The levels of fasting blood glucose (FBG), serum testosterone (T), luteinizing hormone, fasting insulin (FIN) and insulin-like growth factor-1 were also measured, and the homeostasis model assessment of insulin resistance (HOMA-IR) was calculated. The expression level of androgen receptor (AR) in the ovarian tissue, and the cytochrome P450 cholesterol side-chain cleavage enzyme (P450scc) mRNA levels in the ovary and adrenal tissues were detected. The levels of T, FIN, FBG and HOMA-IR in the combination group were significantly reduced; the wet weights of the ovary and the adrenal gland were decreased significantly, while that of the uterus was increased, and the histological morphology benignly recovered. The rats of each treatment group all experienced restored ovulation. The AR expression level in the treatment group was reduced, and the P450scc mRNA levels in the ovary and the adrenal gland of the combined treatment group were decreased. BFPs combined with metformin significantly affected PCOS, and the possible mechanism involved in the treatment may have been through the reduction of P450scc generation. BFPs may reduce the androgen levels, thus allowing the ovary to restore ovulation.

Parkinsons's disease (PD) is a chronic neurodegenerative disorder characterized by the loss of dopaminergic neurons of substantia nigra pars compacta in the ventral midbrain.1)The loss of dopaminergic neurons leads to the reduction of dopamine release into the striatum, and is responsible for the clinical features of PD including bradykinesia, resting tremor, rigidity, and difficulty in initiating movements. 2)Pathological patterns of PD are usually examined by inducing Parkinson's like symptoms in rodents by injection of the dopaminergic neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and is widely used for study of PD. 3)MPTP is converted to MPP ϩ , which structurally resembles dopamine, by brain monoamine oxidase B (MAO-B) and can be actively transported into the dopaminergic nerve terminals of the striatum by dopamine transporter (DAT).4) It is important to note that DAT is found only in dopaminergic neurons and may be the best and only unique marker for dopaminergic neurons.5) It has been shown that administration of MPP ϩ alone decreases dopamine transporter protein expression by Western blot. 6) In addition, MPTP intoxication has been reported to be associated with reduced immunoreactivity of tyrosine hydroxylase (TH), the rate-limiting enzyme of dopamine synthesis, 7) in the dopaminergic neurons of substantia nigra via the dopamine uptake system. This is considered to lead to energy failure and subsequently the inhibition of protein expression.8) The reduction of TH-positive neurons is also considered to be a good marker for dopaminergic cell loss.Current therapy for PD is essentially symptomatic, with the use of levodopa, the direct precursor of dopamine, remaining the primary treatment choice since its first use over 30 years ago.9-11) However, long-term therapy with levodopa is associated with significant side effects.12) Neuroprotective therapy to rescue dopamine neurons, which could alter and prevent the progression of PD, 13) has thus been proposed as an alternative to symptomatic treatment. It has been shown that neuroprotection can be observed in the PD models with iron chelators, radical scavenger antioxidants, MAO-B inhibitors, nitric oxide synthase inhibitors, calcium channel antagonists, glutamate antagonist trophic factors.12) Even nicotine and low exposure to cigarette smoke may have a neuroprotective effect on the dopaminergic nigrostriatal system. 14)In addition, epidemiological data suggest that steroid hormone 17b-estradiol plays an important role in protecting the brain from neurodegenerative processes, including Parkinson's disease. <p class="Abstract">Fundamental to this research was the evaluation of anticoagulant and thrombolytic potential of Berberis orthobotrys using clotting time, prothrombin time and activated partial thromboplastin time performed at 2.5, 5, 10% concentration. Aqueous-methanol extract significantly (p<0.05) increased the coagulation parameters both in human blood as well as in rabbit blood and exhibited the more salient effect on prolongation of these coagulation parameters at 10% concentration. Aqueous methanol extract (25, 50 and 100 mg/kg) produced the preferential increase in bleeding time and clotting time nevertheless the effect was more perceptible after 90 min at 100 mg/kg. Moreover, the crude extract presented a substantial increase in coagulation parameters after 7 days of administration in rabbits with more pronounced effects at the highest dose. HPLC profile of aqueous methanol extract revealed the presence of quercetin, p-coumaric acid, gallic acid, vanillic acid, 4-hydroxy-3-methoxy benzoic acid. Thus, the present study provides the pharmacological basis for its medical use in cardiovascular and other thrombotic disorders.</p>
