-The effect of dietary carbohydrate (CHO) level on serum physiological response, hepatic antioxidant abilities and heat shock protein 70 (HSP70) expression of Wuchang bream (Megalobrama amblycephala) was studied. Two isonitrogenous (28.56% crude protein) and isolipidic (5.28% crude lipid) diets were formulated to contain 30% or 53% wheat starch. Diets were fed for 90 days to fish in triplicate tanks (28 fish per tank). At the end of feeding trial, significantly higher serum triglyceride level, insulin level, cortisol level, and malondialdehyde (MDA) content were observed in fish fed the 53% CHO diet, while significantly lower serum total protein content, alkaline phosphatase activity, superoxide dismutase activity and total antioxidative capacity were found in fish fed the 53% CHO diet compared with those fed the 30% diet. The relative level of hepatic heat shock protein 70 mRNA was significantly higher in the 53% CHO group than that in the 30% CHO at 6, 12 and 48 h after feeding. Ingestion of 53% dietary CHO impacts the nonspecific immune ability and causes metabolic stress in Megalobrama amblycephala.

Effects of astaxanthin on antioxidant capacity of golden pompano (Trachinotus ovatus) in vivo and in vitro

The objective of this research was to study the effect of astaxanthin (AST) on growth performance and antioxidant capacity in golden pompano (Trachinotus ovatus) both in vivo and in vitro. In the in vivo study, two diets were formulated with or without astaxanthin supplementation (D1 and D2; 0 and 200 mg/kg) to feed fish for 6 weeks. In the in vitro study, cells from hepatopancreas of golden pompano were isolated and four treatments with or without astaxanthin and H 2 O 2 supplementation were applied (control group: without both astaxanthin and H 2 O 2 treated; H 2 O 2 group: just with H 2 O 2 treated; H 2 O 2 + AST group: with both astaxanthin and H 2 O 2 treated; AST group: just with AST treated). Results of the in vivo study showed that weight gain (WG) and special growth rate (SGR) significantly increased with astaxanthin supplemented (P < 0.05). Feed conversion ratio (FCR) of fish fed D2 diet was significantly lower than that of fish fed D1 diet (P < 0.05). Hepatic total antioxidant capacity (T-AOC) and the reduced glutathione (GSH) of golden pompano fed D2 diet were significant higher than those of fish fed D1 diet (P < 0.05). Superoxide dismutase (SOD) was significantly declined as astaxanthin was supplemented (P < 0.05). Results of the in vitro study showed that the cell viability of H 2 O 2 group was 52.37% compared to the control group, and it was significantly elevated to 84.18% by astaxanthin supplementation (H 2 O 2 + AST group) (P < 0.05). The total antioxidant capacity (T-AOC) and the reduced glutathione (GSH) of cell were significant decreased by oxidative stress from H 2 O 2 (P < 0.05), but it could be raised by astaxanthin supplementation (H 2 O 2 vs H 2 O 2 + AST), and the malondialdehyde (MDA) was significant higher in H 2 O 2 group (P < 0.05) and astaxanthin supplementation could alleviate the cells from lipid peroxidation injury. In conclusion, dietary astaxanthin supplementation can improve the growth performance of golden pompano. Moreover, astaxanthin can improve the golden pompano hepatic antioxidant capacity both in vivo and in vitro study by eliminating the reactive oxygen species.

We evaluated the effect of vitamin C (Vit C) supplementation on the resistance of Megalobrama amblycephala to Aeromonas hydrophi1a infection under pH stress. Fish were randomly divided into six groups: a control group (fed with a basal diet) and five treatment groups (fed with basal diet supplemented with 33.4, 65.8, 133.7, 251.5 and 501.5 mg/kg Vit C, respectively). After 90 days, fish were exposed to combined stressors, first pH 9.5 followed by A. hydrophila infection. The results showed that 133.7 mg/kg vit C improved complement 4 (C4), anti-superoxide anion free aradical (ASAFER) and heat shock protein (HSP) 70 compared with the control group; and 251.5 mg/kg vit C enhanced complement 3 (C3), HSP60, HSP70 and HSP90 compared to the control group before stress. After pH stress and A. hydrophila infection, hemoglobin, ASAFER and HSP60, HSP70, HSP90 in the groups fed with 133.7 and 251.5 mg/kg vit C were still significantly higher, while serum cortisol in the group fed with 251.5 mg/kg vit C was lower compared to the control group 15 days after pH stress. The cumulative mortality of the control group was higher than that of the five treatment groups at 12, 24 h after A. hydrophila infection. The results of this study suggest that 133.7-251.5mg/kg vit C in a diet could have potential to stimulate immune response, and enhance resistance against high pH stress and A. hydrophila infection of Wuchang bream.
