Effects of acute and chronic aripiprazole treatment on choice between cocaine self-administration and food under a concurrent schedule of reinforcement in rats

Abstract: Rationale-Dopamine D2-like partial agonists such as aripiprazole have received some attention as potential pharmacotherapies for the treatment of psychostimulant addiction. However, the preclinical evaluations so far have focused on acute effects of aripiprazole.Objectives-We tested the hypothesis that aripiprazole, both as acute and as chronic treatment, would preferentially decrease cocaine self-administration while sparing behavior maintained by a natural reinforcer, resulting in a shift in the allocation o… Show more

“…Rate frequency functions were obtained by logarithmically decreasing the frequency of the stimulation pulses from a value that sustained maximal lever pressing to one that failed to sustain lever pressing. studies, showing that Ari counteracts the rewarding/ reinforcing effects of psychostimulants in experimental animals and the subjective and discriminative effects of Amph in humans (Feltenstein et al 2007 ;Lile et al 2005 ;Newton et al 2008 ;Sorensen et al 2008 ;Stoops et al 2006 ;Thomsen et al 2008 ;Wee et al 2007). Ari has been proposed as a potential therapy for psychostimulant dependence (Stoops, 2006) or for the treatment of schizophrenia with comorbid stimulant abuse (Beresford et al 2005).…”

“…Remarkably, a complete and long-term suppression of methamphetamine seeking by food choice was also demonstrated in rats after different histories of extended access to methamphetamine self-administration that are known to induce addiction-like behavior (Caprioli et al, 2015a, b). In contrast, in other choice studies, rats eventually shifted their choice from sweet reward to cocaine when the dose of cocaine was sufficiently high (ie, up to 0.5 mg per injection) (Thomsen et al, 2008(Thomsen et al, , 2013.…”

“…In studies employing a setting where choosing under the drug influence was not possible (eg, by imposing a sufficiently long interchoice interval for drug dissipation before the next choice), most rats preferred sweet reward over cocaine, even at the high doses . In contrast, in studies using a setting where choosing under the drug influence was possible (eg, by not imposing a minimum interchoice interval), virtually all rats preferred the high doses of cocaine (Thomsen et al, 2008(Thomsen et al, , 2013. Thus, it seems that rats choose cocaine when they are under the drug influence at the time of choice and otherwise choose the nondrug reward.…”

“…In other studies, putative D 3 receptor antagonists that decreased cocaine or methamphetamine self-administration did so only at doses that also decreased responding maintained by food (see, eg, Claytor et al, 2006;Martelle et al, 2007;Thomsen et al, 2008;Higley et al, 2011). However, it remains possible that the lack of selective effects on cocaine self-administration in these studies is due to the relatively high D 2 receptor affinity and/or lack of sufficient selectivity for D 3 versus D 2 receptors of the pharmacological tools that were used.…”
