Effect of Peripheral Opioid Receptor Agonists on Depressive Activity of Ethanol

Abstract: We studied the effect of peripheral μ- and κ-opioid receptor agonists (not crossing the bloodbrain barrier) on locomotor activity and metabolism in rats after acute administration of ethanol. Intraperitoneal injection of ethanol in a single dose of 2 g/kg had a strong depressive effect manifested in a decrease in horizontal locomotor activity and suppression of metabolism. μ-Opioid receptor agonist DAMGO and κ-opioid receptor agonist ICI 204,448 partly abolished the effect of ethanol on locomotor activity of r… Show more

“…The first (control) group of animals (n = 10) was administered vehicle alone (water); the second group (n = 10) was administered 200 mg/kg DAMGO (Tocris Bioscience, Bristol, UK); and the third group (n = 10) was administered 200 mg/kg DADLE (Tocris Bioscience). The dose of the administered substances was selected on the basis of our previous experiments (Trigub et al, 2014;Bogdanova et al, 2015).…”

“…The opioid is a substance of synthetic and semi-synthetic origin, which is in its composition, morphochemically, related to opiate receptors and provides a certain psycho-vegetative action. The representative of this group is nalbuphine -a synthetic opioid of an agonist-antagonist group (agonist of k-receptors and an antagonist of mu-receptors) [4,5]. Unfortunately, uncontrolled use of opioids leads to an increase in drug addiction not only in Ukraine, but also in the world [6].…”
