Abstract: Electrostatic interactions play a crucial role in modulating and stabilizing molecular interactions in membranes and membrane-mimetic systems such as micelles. We have monitored the change in the conformation and dynamics of the cationic hemolytic peptide melittin bound to micelles of various charge types, utilizing fluorescence and circular dichroism (CD) spectroscopy. The sole tryptophan of melittin displays a red-edge excitation shift (REES) of 3-6 nm when bound to anionic, nonionic, and zwitterionic micell… Show more

“…The PCR product was cloned into vector pBluescript II KS (pBlue) in the polylinker site with BamHI and SalI, and the resulting sp-H construct was used to generate other chimeric HIV-1 Env mutants. The mellitin SP (the sequence is in Table 2) with a 6-aa linker DPINMT was described previously (22,26), and the corresponding DNA was synthesized through overlapping primer extension by PCR with the primers F mellitin and R mellitin . This mellitin SP coding sequence was cloned into the sp-H construct at XbaI and BamHI sites (pBlue-pre-M).…”

“…The general principle underlying the formation of micelles (i.e., the hydrophobic effect) is common to other related assemblies such as lipid bilayers and biological membranes. Micelles are highly cooperative and are dynamic in nature (31) and have been used as membrane-mimetic systems to characterize membrane proteins and peptides (32).…”

“…2,5 Melittin is intrinsically fluorescent due to the presence of a single tryptophan residue, Trp-19, which makes it a sensitive probe to monitor the interaction of melittin with membranes [6][7][8][9][10][11] and membrane-mimetic systems. 12,13 This is particularly advantageous since there are no other aromatic amino acids in melittin and this makes interpretation of fluorescence data less complicated due to lack of interference and heterogeneity. More importantly, it has been shown that the sole tryptophan residue of melittin is crucial for its powerful hemolytic activity since a dramatic reduction in activity is observed upon photooxidation 14 and substitution by leucine.…”
