The parenteral use of amino acids and vitamins has been used to improve performance in animal production. However, few studies demonstrating the toxicity of these compounds. The present investigation evaluated the toxicological and clinical safety of the injectable supplement compound by amino acids and vitamins, administered by intramuscular route in Wistar rats. 56 females (± 220g), pubescent virgins, were randomly divided into seven groups (n=8): Treated with dose of 0.2, 2 and 4 mL, sampled 24 and 96 hours post-treatment (HPT), totaling six groups and one control group (Physiology Standard). Signifi cant increase in the enzymatic activity of serum ALT, AST, LDH and ALP was observed in animals treated with 2 and 4 mL (24 HPT). However, these changes returned to baseline levels 96 HPT, except in serum AST and ALP in animals treated with 4 mL. CK-NAC showed dose-response effect, since the higher compound dose was followed by proportional increase in the circulating levels of this enzyme, associated to muscle degeneration and necrosis area in rats treat with 4mL at 96 HPT. The evaluation of hepatic function did not result in signifi cant changes in the serum levels of triglycerides, cholesterol, albumin, urea and creatinine in the comparison between rats treated with 0.2 mL and control rats, demonstrating the innocuousness of this treatment on the activity of liver and kidney. However, deleterious effects were observed in animals subjected to doses of 2 and 4 mL, including observed increased splenic and hepatosomatic response, signifi cant decrease in circulating albumin levels and signifi cant leukocytosis, which is characterized by neutrophilia, followed by lymphopenia demonstrating the compound irritating effect on muscle tissue.

RESUMO:Dentre as espécies de peixes que demonstram potencial para o cultivo no Brasil, a tilápia-do-Nilo (Oreochromis niloticus) é considerada a espécie mais produtiva. Essa espécie apresenta diversas vantagens, entre elas: rápido crescimento, maior rendimento de filé, resistência a baixa qualidade de água e boa aceitação no mercado consumidor. Na nutrição de peixes vários são os aspectos a serem avaliados em uma ração. O selênio e a vitamina E fazem parte de um sistema de defesa antioxidante multicomponente. Este sistema protege as células contra os efeitos adversos das espécies reativas de oxigênio e outros radicais livres que iniciam a oxidação. Além disso a suplementação de Vitamina E na dieta proporciona um maior desempenho produtivo, efeitos positivos nos parâmetros imunológicos dos peixes e resistência ao estresse. O selênio é um elemento que possui margem estreita entre os níveis de exigência e toxidez. As exigências são baixas, mas se não atingida, podem comprometer o sistema antioxidante e causar graves consequências para o metabolismo do animal. A capacidade do peixe em acumular selênio nos tecidos pode tornar a carne de pescado um interessante alimento funcional para a nutrição humana, especialmente como boa fonte de selênio. A tilápia-do-Nilo tem um nível de exigência específica de selênio e vitamina E que não pode ser satisfeito somente pelo ambiente aquático. Desta forma uma correta suplementação na dieta de minerais e vitaminas poderá garantir um maior desempenho, proporcionando uma melhor qualidade de carne e melhor aceitação no mercado.Palavras-chave: Antioxidante. Imunológicos SELENIUM AND VITAMIN IN DIETS FOR TILAPIA-DO-NILO SUMMARY:Among the species of fish that demonstrate potential for cultivation in Brazil, Nile tilapia (Oreochromis niloticus) is considered the most productive species. This species has several advantages, including: rapid growth, increased fillet yield, low quality water resistance and good acceptance in the consumer market. In fish nutrition there are several aspects to be evaluated in a ration. Selenium and Vitamin E are part of a multicomponent antioxidant defense system. This system protects the cells against the adverse effects of reactive oxygen species and other free radicals that initiate oxidation. In addition, supplementation of Vitamin E in the diet provides greater productive performance, positive effects on immunological parameters of fish and resistance to stress. Selenium is an element that has a narrow margin between the levels of demand and toxicity. The requirements are low, but if not achieved, could compromise the antioxidant system and cause serious consequences for the animal metabolism. The fish's ability to accumulate selenium in tissues can make fish fillet an interesting functional food for human nutrition, especially as a good source of selenium. RESUMO.-[Anticoagulante Na 2 EDTA danifica amostras de sangue do teleósteo Piaractus mesopotamicus.] O presente estudo teve por objetivo avaliar os efeitos da heparina sódica e Na 2 EDTA no sangue de Piaractus mesopotamicus (360,7±42,4g, 26,4±1,0cm). Foram amostrados vinte peixes em dois ensaios experimentais, sendo dez peixes utilizados para análise da fragilidade dos eritrócitos e dez peixes para análise dos parâmetros hematológicos e estudo bioquímico do plasma. O sangue coletado por punção veno-caudal foi aliquotado e armazenado em diferentes soluções anticoagulantes: Na 2 EDTA 10%, Na 2 EDTA 3%, heparina sódica 5.000 UI e heparina sódica 100 UI. Níveis plasmáticos de cálcio apresentados nas amostras armazenados em Na 2 EDTA diminuíram cerca de 80% em relação aos dois grupos armazenados com heparina. Amostras de sangue de pacus armazenados com Na 2 EDTA demonstraram aumento do hematócrito e VCM, e diminuição na CHCM. O efeito dose-resposta foi observado neste estudo. Estes resultados são reforçados pelos níveis mais elevados de proteína plasmática e hemólise apresentado no sangue armazenado com Na 2 EDTA 10%, o que confirma o efeito deletério desse tratamento anticoagulante na qualidade de amostras de sangue. Na 2 EDTA não é indicada para armazenar amostras de sangue de P. mesopotamicus, e heparina sódica a 100 UI é o anticoagulante mais recomendado, uma vez que este tratamento apresentou a menor taxa de alterações no sangue armazenado. The present study aimed to evaluate the effects of Na heparin and Na 2 EDTA on blood of Piaractus mesopotamicus (360.7±42.4g, 26.4±1.0cm). Twenty fishes were sampled in two experiment trials, ten for erythrocyte fragility analysis and ten for hematologic and plasma biochemical study. The blood collected by venous-caudal puncture was fractioned and stored in anticoagulants solution: Na 2 EDTA 10%, Na 2 EDTA 3%, Na heparin 5000 IU and Na heparin 100 IU. Plasmatic levels of calcium presented in the Na 2 EDTA stored samples were about 80% lower than both heparin groups. Blood samples of P. mesopotamicus stored with Na 2 EDTA demonstrated increase in the hematocrit and MCV, and decrease in MCHC. The dose-response effect was observed in this study. The results are reinforced by the higher levels of plasmatic protein and hemolysis presented in the Na 2 EDTA 10% stored blood, confirming the deleterious effect of this anticoagulant treatment on the quality of blood samples. Na 2 EDTA is not indicated to store P. mesopotamicus blood samples, but sodium heparin at 100 IU is the most recommended anticoagulant, since this treatment presented the lower rate of alterations in the stored blood.
