Abstract: Microbial biofilm formation in dental unit water lines (DUWL) is a phenomenon that has been recognized for nearly four decades. Water delivered by DUWL can harbor high numbers of bacteria, including opportunistic pathogens. Biofilms on tubing within DUWL may serve as a reservoir for these microorganisms and should therefore be controlled. In this study, the effects of eight biocides were monitored on DUWL biofilms individually and in combination by epifluorescence microscopy and total viable counts (TVC). The … Show more

“…FT-IR spectroscopy has been demonstrated to be an effective tool to record the infrared spectrum of whole cells, such as bacteria in a film, or even, the spectrum of a single cell [29]. The six bacterial strains examined for FT-IR spectroscopy were isolated from DUWL biofilm [15]. The results for the spectra indicated that variation in sample preparation has either no/negligible effect on spectra using KBr disc method.…”

“…In addition to the environmental bacteria, dental unit tubings have also been recognized to be contaminated with some opportunistic pathogens [15]. A wide range of organisms has been isolated from DUWL by traditional cultivation techniques and include fungi, free-living amoebae, protozoa, nematodes and human pathogens, such as Pseudomonas aeruginosa, Legionella pneumophila, Mycobacteria species and Staphylococcus species [5].…”

“…Repeated exposure to triclosan, which acts primarily on Gram-positive bacteria, resulted in a slight decrease in the susceptibility of domestic drain biofilm isolate S. maltophilia M9.13 (193). SDS has been assessed for its ability to reduce bacterial biofilms (197). Data from my laboratory indicate that S. maltophilia clinical isolate X26332 survives and persists in a 0.02% solution of SDS for 14 days at 30°C; biofilms of this isolate have been observed to form in Luria-Bertani broth containing 0.02% SDS (22; my unpublished observations).…”

“…Eight biocides (1% sodium dodecyl sulfate, 35% hydrogen peroxide, 5.25% sodium hypochlorite, 1% phenol, 4% Tween 20, 1% EDTA, 0.2% chlorhexidine gluconate, and 1% povidone-iodine) were studied for their effects on biofilm bacteria present in dental unit water lines (197). The flushing of the tubing for 48 h with a combination of 5.25% sodium hypochlorite and 1% phenol resulted in lower biofilm bacterial counts present in the unit water line tubing than in those with flushing with each biocide separately.…”

“…However, for the study of adherent communities, viability tests alone are insufficient (24). The biofilm matrix can be recolonized, and assays should therefore target viable mass and matrix in a discriminatory way (9,12,25). For example, a recently developed S. aureus model uses dimethyl methylene blue (DMMB) for detecting the matrix, while resazurin is used as a viability indicator (24).…”
