Effect of Aminoguanidine in Sperm DNA Fragmentation In Varicocelized Rats: Role of Nitric Oxide

Abstract: The findings of this study suggest that AG improves sperm DNA fragmentation that is associated with infertility in varicocelized rats, and treatment with AG can reduce the damage to sperm DNA.

“…The same deterioration of sperm DNA integrity following varicocele induction in our study could be explained by OS pathologic mechanism. In this study, the finding of negative changes of chromatin structure in varicocele rats as increase in percentage of sperm staining by three methods AB, TB and AO compared to control group is compatible with previous studies (Abbasi et al., ; Bertolla et al., ; Enciso et al., ).…”

“…For stabilizing, the AO stain solution stored at 4°C in a dark bottle for 4 weeks. After staining, the slides were gently rinsed with distilled water and evaluated by a fluorescent microscope (Leitz, Germany) as described in a previous publication (Abbasi et al., ). Same examiner assessed 200 sperms on each slide with an evaluation period less than 40 s per field.…”

“…For intervention studies, some rats ( n = 6) were administered i.p. injections of 30 or 50 mg/kg body weight AG 1 hour prior to the administration of MTX . Some rats ( n = 6) were administered with AG alone 30 or 50 mg/kg body weight intraperitoneally.…”

“…The reference values used were taken from the criteria given by the Guidelines on Male Infertility 2010, provided by the European Association of Urology (EAU). The lower reference limit (with 5th centiles and their 95% confidence intervals) of progressive motility was 0.32 (0.31-0.34) and minimum sperm concentration was 15 Â 10 6 /ml (12)(13)(14)(15)(16) [25].…”

“…Increased levels of NO decrease the intratesticular level of androgens and also inhibit human chorionic gonadotropin hormone (hCG) steroidogenesis [12]. The sperm of infertile men with varicocele exhibit marked DNA damage that appears to be related to high oxidative stress [13][14][15]. The concentration of nitrotyrosine, a biological marker of NO-induced damage, in the spermatic veins is significantly higher than in the peripheral veins of patients with varicocele, indicating that NO causes nitration of plasma proteins with higher molecular weight.…”
