Summary Reef fishes of the order Perciformes represent an interesting group for both genetic and cytogenetic studies as their evolutionary pattern, usually unclear, might range from dynamic to quite stable. A karyotype with 2nϭ48 acrocentric chromosomes is considered basal for this group, whereas karyotypes with other formulae are regarded as derived. Some Perciformes families present a high karyotypic diversification such as Apogonidae, although cytogenetic reports in this family along South Atlantic were absent so far. In the present work, cytogenetic analyses were carried out in Apogon americanus from Northeastern Brazil, revealing a modal diploid number of 2nϭ36 (12mϩ6smϩ16stϩ2a; FNϭ70) and a remarkable presence of large metacentric pairs. The AgNORs were located on short arms of the 8th pair, at telomeric position, coincident with hybridization signals obtained by FISH using 18S rDNA probes. The 5S minor ribosomal genes are distributed in 2 chromosomal pairs, not syntenic to the major ribosomal subunits. Heterochromatic blocks are located mainly at pericentromeric position in most of chromosomal pairs. The early replication pattern showed longitudinal bands with both early and late replication in the heterochromatin segments, being very similar to the pattern obtained after chromosomal digestion with EcoRI restriction enzyme. The present data, coupled with previous reports in other species, suggest a karyotypic evolution mainly driven by Robertsonian fusions (Rb) and pericentric inversions. In spite of the structural rearrangements, some features in the DNA composition and specific regions, such as 5S and 18S rDNA sites, reflect symplesiomorphic karyotype traits, shared by other clades in Perciformes.

Chromosomal evidence of population subdivision in the freshwater fish Leporinus elongatus in the Upper Paraná River basin

Cytogenetic analyses performed in populations of the Neotropical freshwater fish Leporinus elongatus of the Upper Paraná River basin showed a chromosome polymorphism involving heterochromatic segments of the nucleolar organizing pair in this species. The NOR-bearing chromosome pair was characterized by two phenotypes identified by the absence (C 1 ) or presence (C 2 ) of a heterochromatic segment located in an interstitial position of the long arm in this pair. The meiotic segregation of these variations results in three distinct cytotypes, C 1 C 1 , C 1 C 2 and C 2 C 2 . Both populations were in Hardy-Weinberg equilibrium. Sex-related cytotype divergence was identified and multivariate analyses revealed that the C 2 C 2 individuals presented morphometric differentiation relative to body height in relation to the other cytotypes. A chromosome differentiation and a cytotype proportion difference observed between both populations suggest the occurrence of population subdivision within this species along the Upper Paraná River basin. These findings might contribute to the knowledge of the population biology of this fish and for its conservation.

The absence of longitudinal bands in fish chromosomes has been associated with technical problems in chromosome preparations or the absence of a structural compartmentalization in the fish genome. In the present study, a R-banding pattern was obtained using a replication banding technique by in vivo treatment with 5-bromodeoxyuridine (5-BrdU). G-banding patterns were obtained after trypsin treatment and also after chromosome cleavage by in situ treatment with the restriction endonuclease BamHI. A similar G-banding pattern was also obtained after cleavage with the endonuclease HinfI. Presence of a resolute R- and G-banding patterns shows that Astyanax scabripinnis paranae chromosomes could present an isochore-like structure similar to that found in other vertebrates.
A ausência de bandas longitudinais em cromossomos de peixes tem sido associada a problemas técnicos nas preparações cromossômicas ou à ausência de uma compartimentalização estrutural no genoma dos peixes. No presente estudo, um padrão de bandas R foi obtido usando a técnica de bandamento de replicação pelo tratamento in vivo com 5-BrdU. Padrões de bandamento G foram obtidos após tratamento dos cromossomos com tripsina e também após clivagem in situ com a endonuclease de restrição BamHI. Um padrão similar de bandamento G também foi obtido após clivagem dos cromossomos com a endonuclease HinfI. A presença de um resolutível padrão de bandamento R e G em Astyanax scabripinnis paranae mostra que seus cromossomos parecem apresentar estrutura "isochore" similar àquela encontrada em outros vertebrados
