Early replication bands in two scorpion fishes, Scorpaena porcus and S. notata (order Scorpaneiformes)

Abstract: The use of the BrdU-FPG methodology in the scorpion fish (Scorpaena porcus) has allowed us to estimate the mean cell cycle duration in kidney cells in vivo as being approximately 32.5 h. On the basis of this datum, replication protocols from 30 to 10 h of BrdU incorporation were employed revealing a precise order in the replication of the chromosomes. Treatments of 18–20 h proved to be the most useful in getting optimum replication bands and were used to elaborate the RBG-banding pattern karyotypes of S. porcu… Show more

“…The specimens were submitted to mitotic stimulation as proposed by Molina (2001) for a period of 24-48 h. Chromosomal preparations were obtained from kidney tissue according to Gold et al (1990). About 30 metaphases were analyzed per specimen.…”

“…Early replication bands were obtained by in vivo incorporation of 5Ј-Bromo-2Ј-deoxyuridine (5ЈBrdU), following the methodology described by Giles et al (1988). The base-analogous 5ЈBrdU (5 mg/ml in 0.9% NaCl solution) was injected in the specimens via intraperitoneal at a proportion of 1 ml/100 g of body weight for a period of 6 1/2 h prior to cytogenetic preparations.…”

“…Replication banding patterns have been obtained for several fish species (Delany and Bloom, 1984;AlmeidaToledo et al, 1988;Giles et al, 1988;Gold et al, 1990;Hellmer et al, 1991;Sánchez et al, 1993;Bertollo et al, 1997) after brief treatment with BrdU. A replication banding pattern was obtained in chromosome preparations of A. scabripinnis paranae using this technique (Figures 1a and 2a).…”
