Abstract: Rubisco, the key enzyme of CO2 fixation in photosynthesis, is prone to inactivation by inhibitory sugar phosphates. Inhibited Rubisco undergoes conformational repair by the hexameric AAA+ chaperone Rubisco activase (Rca) in a process that is not well understood. Here we performed a structural and mechanistic analysis of cyanobacterial Rca, a close homolog of plant Rca. In the Rca:Rubisco complex, Rca is positioned over the Rubisco catalytic site under repair and pulls the N-terminal tail of the large Rubisco s… Show more

“…RCA: structure, regulation, and its thermo-lability RCA is a member of the AAA+ ATPases (ATPases associated with diverse cellular activities), displaying a core twodomain architecture across prokaryotic and eukaryotic RCA, with a flexible α/β-nucleotide binding subdomain at the N-terminus and an α-helical subdomain at the Cterminus (Shivhare and Mueller-Cajar 2017b;Flecken et al 2020). RCAs of plants and green algae also contain a Rubisco recognition domain N-terminal to the AAA+ core while the ATPase activity is located near the extension at its C-terminus, which is critical for its activity as shown by the loss of function mutant analyses (Bhat et al 2017b).…”

“…However, deciphering the factors involved in driving these regulatory activities and their interaction in this complex network, requires further exploration. With the identification and characterization of RCA-like proteins in lower organisms such as cyanobacteria might shed some light in our understanding of the importance of activase and its evolution towards its role in Rubisco activation (Flecken et al 2020). Further, in evaluating the effects of overexpressing RCA, the developmental stage of leaves as well as the ratio of Rubisco vs RCA appear to be important in achieving the optimal photosynthetic efficiency.…”

“…The core AAA + module consists of a nucleotide-binding α/β sub-domain (from helix H 0 to H 5 ) and α helical sub-domain (i-helix H 6 -H 10 ) (Stotz et al, 2011;Hasse et al, 2015;Flecken et al, 2020).…”

“…5-17c). Notably, these crosslinked complexes appeared to resemble the CbbX-CbbLS interaction model (Bhat et al, 2017b) as well as the recently described plant/cyanobacteria Rca-ECMC model (Flecken et al, 2020a;Ng et al, 2020), suggesting that Rca2 may also adopt a side-on geometry in regard to its interaction with Rubisco. In addition, observed Rca2-Rubisco complexes were homogenous, with each containing a single Rca2 hexamer per Rubisco hexadecamer.…”

“…Collection of data at a defined tilt was anticipated to resolve this issue (Tan et al, 2017). However, prior to further optimization for data collection, an exquisite structural snapshot of a prokaryotic carboxysome-associated green-type Rca hexamer bound to cyanobacterial Rubisco was communicated on the Biorxiv pre-print server (Flecken et al, 2020a;Flecken et al, 2020b). Consistent with our findings, the study reports of the prokaryotic Rca core (without the C-terminal SSUL domain) adopting a spilt washer conformation that threads the RbcL N-terminus of an inhibited cyanobacterial Rubisco complex through the axial pore.…”
