Bottle gourd (Lagenaria siceraria) is an important vegetable crop as well as a rootstock for other cucurbit crops. In this study, we report a high-quality 313.4-Mb genome sequence of a bottle gourd inbred line, USVL1VR-Ls, with a scaffold N50 of 8.7 Mb and the longest of 19.0 Mb. About 98.3% of the assembled scaffolds are anchored to the 11 pseudomolecules. Our comparative genomic analysis identifies chromosome-level syntenic relationships between bottle gourd and other cucurbits, as well as lineage-specific gene family expansions in bottle gourd. We reconstructed the genome of the most recent common ancestor of Cucurbitaceae, which revealed that the ancestral Cucurbitaceae karyotypes consisted of 12 protochromosomes with 18 534 protogenes. The 12 protochromosomes are largely retained in the modern melon genome, while have undergone different degrees of shuffling events in other investigated cucurbit genomes. The 11 bottle gourd chromosomes derive from the ancestral Cucurbitaceae karyotypes followed by 19 chromosomal fissions and 20 fusions. The bottle gourd genome sequence has facilitated the mapping of a dominant monogenic locus, Prs, conferring Papaya ring-spot virus (PRSV) resistance in bottle gourd, to a 317.8-kb region on chromosome 1. We have developed a cleaved amplified polymorphic sequence (CAPS) marker tightly linked to the Prs locus and demonstrated its potential application in marker-assisted selection of PRSV resistance in bottle gourd. This study provides insights into the paleohistory of Cucurbitaceae genome evolution, and the high-quality genome sequence of bottle gourd provides a useful resource for plant comparative genomics studies and cucurbit improvement.

Breeding melon for resistance to Fusarium wilt: recent developments

Melon Fusarium wilt (MFW), caused by Fusarium oxysporum f. sp. melonis (Fom), is one of the most destructive diseases of melon (Cucumis melo L.). The development and deployment of resistant cultivars is generally considered to be the best approach to control MFW. Based on the host resistance genes associated with variants of this pathogen, Fom isolates were classified into four physiological races designated 0, 1, 2, and 1,2. Two dominant resistance genes, Fom-1 and Fom-2, control resistance to races 0 and 2, and 0 and 1, respectively. Fom isolates classified as race 1,2 are able to induce disease in melon lines carrying the above resistance genes. Many sources of resistance to Fom races 0, 1, and 2 have been reported. Partial resistance to race 1,2 controlled by polygenic recessive genes was only detected in a few Far Eastern melon accessions, except for the breeding line BIZ where complete resistance was described. Identification of DNA markers tightly linked to genes conferring resistance to Fom has immediate application in MFW resistance breeding programs. The Fom-2 gene has been cloned, and it encodes a protein with a nucleotide binding site (NBS) and leucine-rich repeats domain (LRR). Based on the sequence of this domain, some molecular markers linked to this gene were developed. Several DNA markers linked to Fom-1 have also been described. However, the usefulness of these markers was variety-dependent. Therefore, their combined use would be very useful in marker assisted selection for introducing resistance to Fom races 0 and 2 in melon. Recently, these markers were used for the positional cloning of this gene, which encoded a protein with a NBS–LRR domains that shows similarity to the toll and interleukin-1 receptores (TIR). Regarding Fom race 1,2, nine QTL were detected on five linkage groups by composite interval mapping. In this paper we review the current knowledge of MFW disease, and focus on genetic resistance to Fom and marker-assisted selection for resistance.

The availability of the genome sequence of many crop species during the past few years has opened a new era in plant biology, allowing for the performance of massive genomic studies in plant species other than the classical models Arabidopsis and rice. One of these crop species is melon (Cucumis melo), a cucurbit of high economic value that has become an interesting model for the study of biological processes such as fruit ripening, sex determination and phloem transport. The recent availability of the melon genome sequence, together with a number of genetic and genomic resources, provides powerful tools that can be used to assist in the main melon breeding targets, namely disease resistance and fruit quality. In this review, we will describe recent data obtained combining the use of a melon near isogenic line (NIL) population and genomic resources to gain insight into agronomically important traits as fruit ripening, resistance to Cucumber Mosaic virus (CMV) and the accumulation of sugars in fruits.
