Drug-resistant epimutants exhibit organ-specific stability and induction during murine infections caused by the human fungal pathogenMucor circinelloides

Abstract: The environmentally ubiquitous fungus Mucor circinelloides is a primary cause of the emerging disease mucormycosis. Mucor infection is notable for causing high morbidity and mortality, especially in immunosuppressed patients, while being inherently resistant to the majority of clinically available antifungal drugs. A new, RNAi-dependent, and reversible epigenetic mechanism of antifungal resistance – epimutation - was recently discovered in M. circinelloides. However, the effects of epimutation in a host-pathog… Show more

“…Male OF-1 mice weighing 30 g and two months old (Charles River, Barcelona, Spain) [ 30 , 32 , 33 ] were used as animal models for virulence assays. The mice were immunosuppressed by intraperitoneal administration of cyclophosphamide (200 mg/kg of body weight), 2 days prior to infection and then once every 5 days.…”

“…The mice were immunosuppressed by intraperitoneal administration of cyclophosphamide (200 mg/kg of body weight), 2 days prior to infection and then once every 5 days. Eight-mouse groups were challenged intravenously by retroorbital injection of 1 × 10 6 spores [ 32 ]. The assay was repeated twice using two independent mutants of mcwc-1a (MU242 and MU243).…”

“…After discovering the antifungal resistance against FK506 by the epimutational pathway, the activity of this new mechanism was examined in other target genes and antifungal compounds [54,55]. This study analized the resistance to the antifungal compound 5-fluoroorotic acid (5-FOA) by specific silencing of the genes pyrF and pyrG.…”

“…The epimutant generation and antifungal drug resistance processes were also studied using a murine in vivo model [55]. In this model, mice were infected with epimutant strains resistant to the antifungal agent FK506.…”
