Double‐stranded RNA targeting calmodulin reveals a potential target for pest management of Nilaparvata lugens

Abstract: NlCaM plays important roles mainly in nymph development and uptake of vitellogenin by ovaries in vitellogenesis in N. lugens. dsRNA derived from the less conserved 3'-UTR of NlCaM shows great potential for RNAi-based N. lugens management. © 2018 Society of Chemical Industry.

“…RNAi bioassay was performed by injection as previously reported ( Wang et al, 2018 ). Briefly, 200 ng (0.05 μL) and 400 ng (0.1 μL) dsRNA (concentration estimated 4.000 mg/mL) and the same amount of ds GFP (negative control) were injected into the newly emerged fourth- and fifth-instar nymphs (N4D1 and N5D1; Xu et al, 2015 ).…”

“…Therefore, the selection of the target to be silenced is a crucial element. Previously, silencing of genes involved in the metabolism of N. lugens , by injection or feeding of dsRNAs, has shown the potential of RNAi based pest control (Chen et al, ; Ge et al, ; Li et al, ; Wang, Wan, Lai, Zhu, & Fu, ).…”

“…For example: In second-instar Euschistus heros , 27.4 ng dsRNA injection reduced the EhATPase A and Ehact-2 transcript levels by 90% and 59%, respectively, (Castellanos et al, 2018). In third-instar Nilaparvata lugens nymphs, the transcript levels of NlCaM and NlCaM1 at 4 days after injection of 50 ng dsRNA were reduced by 67.9% and 62.8%, respectively, (Wang et al, 2018). Finally, in Nephotettix cincticeps , the expression levels of NcSP75 had decreased to 0.2–0.5% of those of the control group at 2, 4, and 8 d after a 9 ng dsRNA-injection (Matsumoto and Hattori, 2018).…”
