“…The phosphoinositide 3-kinase (PI3K) blocker 3-MA is routinely used as an inhibitor of autophagy 34 , and markedly abrogated the effects of PD on apoptosis and ROS generation. The expression levels of inflammation cytokines (IL-6, TNF- α, and MMP-13) and de-differentiation markers (Col I) were also significantly increased by treatment with 3-MA, this result is consistent with the findings of autophagy protection in existing researches 3537 , one possible explanation is that induction of autophagy plays the key role in maintaining the phenotype of chondrocyte and inhibiting inflammatory response. In short, our findings reveal that PD likely relieves symptoms by activating autophagy, however, more studies are required to clarify the further mechanism.…”

“…In comparison, in the treatment of rheumatoid arthritis (RA), fumagillin prodrug (Fum-PD) NPs can promote autophagic flux, followed by inhibition of the NF-κB signaling pathway-dependent inflammatory response [232]. Additionally, based on the significant effectiveness of anti-ROS therapy, dopamine melanin NPs were utilized to further decelerate cartilage degradation through autophagy activation for efficient treatment of osteoarthritis (OA) [233]. Subsequently, the targeted regulation of autophagy by using direct NMs or as drug carriers has introduce huge potential for clinical treatment of related diseases.…”

“…The production of ROS by dye 847-ZnO NPs was measured using NBT and DCFDA assay. The NBT assay is a colorimetric assay for ROS production in which water-soluble NBT salt is converted to water-insoluble, blue NBT-diformazan in the presence of ROS [65][66][67][68]. DCFDA is commonly used to detect ROS in cells by oxidizing the non-fluorescent molecule to fluorescent 2 ,7 -dichlorofluorescein (DCF); the amount of which is directly proportional to the ROS present [67][68][69].…”

DCFDA is commonly used to detect ROS in cells by oxidizing the non-fluorescent molecule to fluorescent 2 ,7 -dichlorofluorescein (DCF); the amount of which is directly proportional to the ROS present [67][68][69]. The light source used in this study to induce ROS production from the Dye 847-ZnO NPs was a NIR LED lamp (λ = 850 nm, Power density = 4.6 mW/cm 2 ).…”
