“…Incorporation of [3H]thymidine is a valid technique for estimating DNA synthesis and cell proliferation because a close agreement is obtained between changes in the incorporation of [3H]thymidine and variations in the activity of DNA polymerase (Jahn, Kalbermann, Machiavelli, Szijan & Burdman, 1980) and the mitotic index (Lloyd, Jacobi & Meares, 1978). In the present study we have shown that the inhibition of production of prolactin by the pituitary gland in the tumour-bearing hyperprolactinaemic rat is accompanied by a marked decrease in the synthesis of pituitary DNA.…”

“…In our present study the steroid was capable of restoring prolactin production and DNA synthesis in the suppressed pituitary glands of hyperprolactinaemic tumour-bearing rats. Conversely, we previously reported that pituitary enlargement produced by chronic treatment with oestrogen was significantly reversed by the administration of bromocriptine, a dopaminergic agent (Kalbermann et al 1980). Under these circumstances, there was also a considerable decrease in DNA synthesis, activity of DNA polymerase a, and a reduction in serum prolactin levels with a concomitant accumulation of prolactin within the gland.…”

“…Thus, it is accompanied by higher activity of the enzyme DNA polymerase-, which is the replicative enzyme. In several mammalian systems including the anterior pituitary gland, the activity of this enzyme changes in parallel with variations in DNA replication (Kalbermann, Szijan & Burdman, 1919a;Jahn et al 1980). On the other hand, the activity of DNA polymerase-ß, which is the reparative enzyme, did not change after castration (data not shown).…”

“…The reaction mixture was incubated at 37°C for 15 and 30 min. The reaction was stopped and the radioactivity measured as described previously (Jahn et al 1980). The DNA polymerase-activity was characterized using the specific inhibitor V-ethylmaleimide (Sigma) at the optimum pH for both the Tris-soluble and the phosphate-soluble fractions.…”
