Flax dew-retting is a key step in the industrial extraction of fibers from flax stems and is dependent upon the production of a battery of hydrolytic enzymes produced by micro-organisms during this process. To explore the diversity and dynamics of bacterial and fungal communities involved in this process we applied a high-throughput sequencing (HTS) DNA metabarcoding approach (16S rRNA/ITS region, Illumina Miseq) on plant and soil samples obtained over a period of 7 weeks in July and August 2014. Twenty-three bacterial and six fungal phyla were identified in soil samples and 11 bacterial and four fungal phyla in plant samples. Dominant phyla were Proteobacteria, Bacteroidetes, Actinobacteria, and Firmicutes (bacteria) and Ascomycota, Basidiomycota, and Zygomycota (fungi) all of which have been previously associated with flax dew-retting except for Bacteroidetes and Basidiomycota that were identified for the first time. Rare phyla also identified for the first time in this process included Acidobacteria, CKC4, Chlorobi, Fibrobacteres, Gemmatimonadetes, Nitrospirae and TM6 (bacteria), and Chytridiomycota (fungi). No differences in microbial communities and colonization dynamics were observed between early and standard flax harvests. In contrast, the common agricultural practice of swath turning affects both bacterial and fungal community membership and structure in straw samples and may contribute to a more uniform retting. Prediction of community function using PICRUSt indicated the presence of a large collection of potential bacterial enzymes capable of hydrolyzing backbones and side-chains of cell wall polysaccharides. Assignment of functional guild (functional group) using FUNGuild software highlighted a change from parasitic to saprophytic trophic modes in fungi during retting. This work provides the first exhaustive description of the microbial communities involved in flax dew-retting and will provide a valuable benchmark in future studies aiming to evaluate the effects of other parameters (e.g., year-to year and site variability etc.) on this complex process.

To explore high-quality microbial resources with the capability of ramie degumming, we collected soil samples from rotten ramie and straw heaps. After enrichment culture by ramie raw materials, bacterial strains with the potential ramie-degumming function were screened using a pectin-hydrolysis plate. Dominant bacteria were identified by combining colonial morphological characteristics with the molecular biology method, and their ramie-degumming effects were verified through comprehensive biological degumming indices. Results demonstrated that Bacillus aryabhattai, Bacillus thuringiensis, Lysinibacillus fusiformis, and Acidovorax temperans were successfully obtained. The highest pectinase activity, 98.2 U/mg, was found by A. temperans. B. thuringiensis showed the best ramie-degumming effect. The residual gum content, single-fiber linear density, and bundle-breaking strength of the degummed ramie fiber treated with B. thuringiensis were 8.32%, 6.80 dtex, and 7.84 cN/dtex, respectively. The residual gum content of the ramie fiber treated with B. thuringiensis met the textile requirement (<10%), and the values of all other indicators were also satisfactory. Therefore, B. thuringiensis was an excellent strain for ramie degumming, indicating potential industrial applications.

The purpose of this study was to use a historical series of exsiccates as a documentary source, aiming to retrieving information on species of Urticaceae Juss. The study approached the collections of exsiccates belonging to herbaria of the Royal Botanic Gardens (K), Kew, England; Muséum National d’Histoire Naturelle (P), Paris, France; and New York Botanical Garden (NY), New York, USA, also printed sources and manuscripts. We analyzed more than 2800 exsiccates, comprising the period between 1783 and 2003. From the information labels of the exsiccates, it was possible to obtain the use and common name of the Urticaceae species. A total of thirteen species with register of use were find in the labels, six genera of Urticaceae: five species of Pourouma Aubl., three of Cecropia Loefl., two of Urera Gaudich., one of Boehmeria Jacq., one of Coussapoa Aubl., one of Laportea Gaudich. Five categories of usage were identify: (1) animal feeding; (2) human feeding; (3) medicinal; (4) fiber and (5) fuel. The analysis of the labels registers pointed to be efficient to raise information related to Urticaceae species, and combined to the research of printed sources and manuscripts provide more details to the data. Furthermore, adding information to the review of the collections history in Brazil. It is necessary to establish initiatives which facilitate the access to documents associated to exsiccates, like the manuscripts of collectors, for continuing the advances in the retrieval of knowledge registered.
