Distance-dependent regulation of NMDAR nanoscale organization along hippocampal neuron dendrites

Abstract: Hippocampal pyramidal neurons are characterized by a unique arborization subdivided in segregated dendritic domains receiving distinct excitatory synaptic inputs with specific properties and plasticity rules that shape their respective contributions to synaptic integration and action potential firing. Although the basal regulation and plastic range of proximal and distal synapses are known to be different, the composition and nanoscale organization of key synaptic proteins at these inputs remains largely elusi… Show more

“…We sought to confirm whether the quality of the images was sufficient to extract biologically relevant features (Methods). For both proteins, the measured quantitative features matched with values previously reported in the literature, enabling the resolution of the 190 nm periodicity of the F-actin lattice in axons, and the size distribution of CaMKII-nanoclusters [53,54,56] (Figure 6a and and Extended Figure 3). Next, we wanted to validate that the agent would adapt it's parameter selection to structures, fluorophores properties or imaging conditions that were not included in the training set.…”

“…Localization of inputs onto dendrites determine dendritic computations and can confer special properties that are important for the function of the neuron in a circuit (Spruston, 2008; Gidon & Segev, 2012; Ferreira et al, 2020). In terms of connectivity, with respect to LC4 and LPLC2, LC4 contacts medial GF dendritic regions, and LPLC2 to lateral regions.…”

“…Thus, we speculated that the effects of anti-NMDAR encephalitis on the brain are asymmetrical. This might be because NMDARs are unevenly distributed on both sides of the brain ( Ferreira et al, 2020 ). For a right-handed person, the left hemisphere is their dominant hemisphere, responsible for language production and comprehension, motor processing, and logical interpretation ( Rentería, 2012 ).…”
