Discovery of 1,2,3-Triazole Derivatives for Multimodality PET/CT/Cryoimaging of Myelination in the Central Nervous System

Abstract: Myelin pathology is present in many neurological conditions such as multiple sclerosis (MS) and traumatic spinal cord injury (SCI). To facilitate development of novel therapies aimed at myelin repair, we set out to develop imaging agents that permit direct quantification of myelination in vivo. In this work, we designed and synthesized a series of fluorescent fluorinated myelin imaging agents that can be used for in vivo positron emission tomography (PET) imaging combined with subsequent post-mortem fluorescen… Show more

“…Additionally, diffusion MRI protocols such as diffusion tensor imaging (DTI) or neurite orientation dispersion and density imaging (NODDI) 13 allow the investigation of microstructural alterations (mainly white matter damage) and discrimination of CSF, extracellular and www.nature.com/scientificreports/ intracellular water. The results obtained with perfusion (DSC-MRI), permeability (DCE-MRI) or diffusion (DTI and NODDI) protocols could be accurately validated with our technique, by means of the co-registration of MRI with high-resolution confocal images obtained upon in vivo labeling of vessels (already explored in this work) 40 .…”

“…Myelin integrity is an important factor in maintaining the physiological function of the central nervous system. e preservation and regeneration of myelin integrity after SCI plays a key role in the recovery of spinal cord function [18,19]. Acute disturbance of nodal organization after SCI exposes the axon and triggers conduction block in the absence of overt demyelination.…”
