Differentiation of murine embryonic stem cells induces progesterone receptor gene expression

Abstract: The role of steroid hormone receptors in very early embryonic development remains unknown. Clearly, expression during organogenesis is important for tissue-specific development. However, progesterone receptor (PR) and estrogen receptors (ERα, ERβ), are expressed during early development through the blastocyst stage in mice and other species, and yet are not essential for embryonic viability. We have utilized the mouse embryonic stem (mES) cell model to investigate the regulated expression of these receptors du… Show more

“…The last set of genes, termed "OFF," were not detectable during the early stages of differentiation examined in these studies. Afp (endoderm) and ␤H1 globin (later mesoderm) did not increase significantly until after 5 days of LIF withdrawal (6). Prl and Ins are tissue-specific genes that were not expressed after LIF withdrawal in our model system even after longer induction times.…”

“…PR is a complex gene, which we have recently shown is turned on in mES cells 12-24 h after LIF withdrawal ( Fig. 1) (6). Two isoforms of PR, PRA and PRB, are encoded by a single gene with two separate promoters (18).…”
