Abstract: Human embryonic stem cells (hESCs) have the ability to differentiate into all the three lineages and are an ideal starting material to obtain cells of desired lineage for regenerative medicine. Continued efforts are needed to evolve more robust protocols to obtain cells of desired lineages and in larger numbers. Also, it has now been realized that rather than transplanting fully committed cells differentiated in vitro, it may be ideal to transplant committed progenitors which retain the intrinsic ability to pr… Show more

“…Initial work from our group resulted in the derivation of two hES cell lines KIND1 and KIND-2 3 , we studied their propensity 36 , adapted both the cell lines to feeder-free conditions and established directed differentiation protocols to make pancreatic 37 and tripotent cardiovascular progenitors 38 . At present, pre-clinical evaluation of safety, efficacy and feasibility of these progenitors is being studied in animal models.…”

“…The threshold values (Ct) were obtained from CFX 96 manager software (Bio-Rad) and normalized using housekeeping gene GAPDH. The primer sequences 12 14 15 are given in Table I . The amplification conditions comprised initial denaturation at 95°C for 5 min, followed by 40 cycles of denaturation at 95°C for 10 sec, annealing at 62°C for 20 sec and elongation at 72°C for 30 sec.…”

“…However, cellular therapy based on CNS-derived neural stem cells has encountered many restrictions and difficulty of use in a clinical situation, due to their limited expansion ability in culture. In fact, while embryonic stem cells are totipotent, and have retained the ability to differentiate into all animal tissues, it is believed that adult stem cells have the limited ability to differentiate only into the cells of the tissue in which they reside [ 22 – 27 ].…”
