Differential regulation of miR-21 and miR-146a by Epstein–Barr virus-encoded EBNA2

Abstract: The discovery of microRNA (miR) represents a novel paradigm in RNA-based regulation of gene expression and their dysregulation has become a hallmark of many a tumor. In virally associated cancers, the host–pathogen interaction could involve alteration in miR expression. Epstein–Barr virus (EBV)-encoded EBNA2 is indispensable for the capacity of the virus to transform B cells in vitro. Here, we studied how it affects cellular miRs. Extensive miR profiling of the virus-infected and EBNA2-transfected B lymphoma c… Show more

“…MiR-222, miR-223, the miR-17~92 cluster, and miR-155 are associated with the differentiation of T cells (Riley et al, 2012;Poole and Sinclair, 2015;Saki et al, 2015). EBV-encoded EBNA2 induces the expression of miR-21, consequently promoting B cell transformation and driving the antiviral immune response (Rosato et al, 2012;Zhu et al, 2013). The upregulation of miR-127 in EBV-positive Burkitt's lymphoma may cause the downregulation of the B cell markers, B lymphocyte-induced maturation protein 1 (BLIMP-1) and X-box binding protein 1 (XBP-1) (Onnis et al, 2012).…”

“…EBV LMP1, which contributes to cell transformation, has been shown to affect the levels of induction of several miRNAs, including miR-146a (37). Similarly, the EBV latency protein EBNA2 alters the levels of some cellular miRNAs, in particular, upregulating miR-21 and downregulating miR-146a (38). Cellular miRNAs can also affect the balance between EBV latent and lytic infection.…”
