Differential gene expression in tumorigenic and nontumorigenic HeLa normal human fibroblast hybrid cells

Abstract: Fusion of tumorigenic HeLa cells with human skin fibroblasts results in chromosomally stable hybrids that are nontumorigenic and no longer express the HeLa tumor-associated marker intestinal alkaline phosphatase (IAP). Previous studies of spontaneous tumorigenic segregants from the nontumorigenic hybrids implicated the loss of one copy of human fibroblast chromosome 11 in the concomitant reexpression of tumorigenicity. In an attempt to identify genes involved in the control of tumorigenic expression, we perfor… Show more

“…A second independent observation from studies of somatic cell hybrids between normal fibroblasts and HeLa carcinoma cells identified Fap as a potential inhibitor of tumorigenesis (Tsujimoto et al, 1999). Fap was one of eight genes differentially expressed in nontumorigenic hybrid lines, consistent with the hypothesis that Fap is involved in suppressing the tumorigenic phenotype.…”

Exaggerated anticipatory anxiety is common in social anxiety disorder (SAD). Neuroimaging studies have revealed altered neural activity in response to social stimuli in SAD, but fewer studies have examined neural activity during anticipation of feared social stimuli in SAD. The current study examined the time course and magnitude of activity in threat processing brain regions during speech anticipation in socially anxious individuals and healthy controls (HC). Method Participants (SAD n = 58; HC n = 16) underwent functional magnetic resonance imaging (fMRI) during which they completed a 90s control anticipation task and 90s speech anticipation task.

“…These indicated that Axl might be related to the tumorigenesis and tumor progression. Sgk, a serine/threonine protein kinase, was found up-regulation in the tumorigenic HeLa cells compared to nontumorigenic HeLa cells which came from fusion of tumorigenic HeLa cells with human skin fibroblasts [55] . Male germ cellassociated kinase (Mak) was shown to be up-regulated in prostate cancer cell lines than those of normal prostate epithelial cells [56] .…”

“…Ramirez‐Montagut et al observed that FAP expression or a catalytic mutant of FAP might decrease the tumorigenicity of mouse melanoma cells in animals and restored contact inhibition and growth factor dependence. Tsujimoto et al identified FAP as a potential inhibitor of tumorigenesis of HeLa carcinoma cells. Taken together, there is an obvious discrepancy between FAP function in tumor promotion and tumor suppression.…”
