Differential display-mediated identification of three drought-responsive expressed sequence tags in tea [Camellia sinensis (L.) O. Kuntze]

Abstract: There is no information on drought-modulated gene(s) in tea [Camellia sinensis (L.) O. Kuntze], a woody and perennial plant of commercial importance. Using differential display of mRNA, three drought-modulated expressed sequence tags (ESTs) were identified. Northern and BLAST analysis revealed that clone dr1 (drought-responsive), induced only by drought but not by ABA, showed significant scores with PR-5 (pathogenesis related) family of PR-protein gene. Another clone dr2, repressed by drought but not by ABA, h… Show more

“…A better understanding of the genetic basis of stress tolerance in tea is essential for enhancing the efficiency and effectiveness of tea breeding programmes. In relation to drought, dr1, dr2 and dr3 drought responsive ESTs (expressed sequence tags) from leaf tissues have been reported using the differential display technique (Sharma and Kumar 2005). In addition, 572 ESTs from young roots of drought tolerant tea cultivars have been introduced using the suppression subtractive hybridization (SSH) technique (Das et al 2012).…”

“…Drought stress (DS) was created by withholding water during the entire period of experimentation. Abscisic acid (ABA, 100 lM) and gibberellic acid (GA 3 , 50 lM) were sprayed separately and also applied to the soil (Sharma and Kumar 2005). AB and the 1st leaf combined were harvested and used for various analyses.…”

“…The genetic mechanism of adaptive responses to drought stress in plant has also been reviewed. Drought-modulated genes (dr1,dr2 and dr3) have also been identified in Camellia sinensis L. (O) Kuntze (Sharma and Kumar 2005). Drought stress caused imbalance between the generation and quenching of reactive oxygen species (ROS).…”
