Nigella sativa or black seed is used as a medicinal plant around the globe. Oil and seeds have a long tradition of folklore use in various medicinal and food systems. The conventional therapeutic use of Nigella sativa, in different ways, has been reported in several studies to treat different diseases including influenza, headache, hypertension, diabetes, inflammation, eczema, fever, cough, asthma, bronchitis, and fever. Based on previously reported potential therapeutic uses of N. sativa compounds, and keeping in mind the dire need of time for the development of potent antiviral, a combined docking, ADMET properties calculation, molecular dynamics, and MM-PBSA approaches were applied in the current study to check the therapeutic potentials of N. sativa chief constituents against COVID-19. Among the studied compounds, we found that dithymoquinone (DTQ), with binding affinity of À8.6 kcal/mol compared to a positive control (chloroquine, À7.2 kcal/mol) , has the high potential of binding at SARS-CoV-2:ACE2 interface and thus could be predicted as a plausible inhibitor to disrupt viral-host interactions. Molecular dynamics simulation of 100 ns well complemented binding affinity of the compound and revealed strong stability of DTQ at the docked site. Additionally, MM-PBSA also affirms the docking results. Compound DTQ of the present study, if validated in wet lab experiments, could be used to treat COVID-19 and could serve as a lead in the future for development of more effective natural antivirals against COVID-19.

Background: Parasitic infections are a major difficulty in tropical and subtropical countries. Traditionally medicinal plants have been used in folk medicine to treat parasitic infections and are a valuable source of novel anti-parasitics. Objective: In our search for therapeutic alternatives to anti-protozoal chemotherapy, thymoquinone, the active ingredient of Black cumin (Nigella sativa) was examined. Materials and Methods: Thymoquinone was tested against Entamoeba histolytica and Giardia lamblia using in vitro susceptibility assays and the mortality of the parasites were then obtained using the standard calculations. The compound was also tested for 48 and 72 hours on both parasites. Results: The current study indicate that the mortality of TQ showed 85.5%, 91.5% and 96.8% mortality on E. histolytica for 25 ppm at 24 hr, 48 and 72 hr, respectively, with IC 50 2¥10-19 ,. On the other hand, this natural compound showed a mortality of 82.83%, 91.76% and 96.62% mortality on G. lamblia for 25 ppm at 24 hr, 48 and 72 hr, respectively, with IC 50 4.8¥10-5. Metrondizole powder gave 70.9% mortality at 156 ppm at the same times. Conclusion: The current results indicate that TQ is more potent on E. histolytica compared to G. lamblia. Further pharmacological studies were needed to help in the clinical presentation of thymoquinone.

COVID-19 has reached to an unimaginable height with several heterogeneous affect on the society and on our regular lives with respect to personal, social, mental, emotional, and economical parameters. To date, most of its physiognomies are not well-known to scientists and researchers. Several studies are being developed round the year to clearly understand the features of the disease. In addition, the lack of authentic information regarding corona virus has led to misinformation and myths in the society. However, myths and misconceptions do not remain myths when people start believing those by hearts despite any strong scientific backing. This study explores such trendy myths with possible arguments and facts. Myths widespreaded in the developing countries as well as mentioned by the World Health Organization (WHO) are critically analysed in this study. Precisely, the study also reveals the risks or gains attained by practicing those myths to fight COVID-19 pandemic. Total 17 COVID-19 related myths have been critically analysed to infer 3 major hypotheses by this study. Finally, the study concludes that legal steps and mass awareness campaigns against or for those myths must be introduced immediately to combat corona virus outbreak. To mention that, when the paper is being written, April-May, 2020, the world is suffering by the devastating Covid-19 pandemic as well as no vaccine or medicine has yet been invented. Furthermore, the paper may help the fellow researchers to deal with the similar diseases with similar symptoms.
