Dietary Pterostilbene and Resveratrol Modulate the Gut Microbiota Influenced by Circadian Rhythm Dysregulation

Abstract: Scope A causal relationship between circadian misalignment and microbiota dysbiosis has been discussed recently, due to their association to pathogenesis. Herein, the possible impact of pterostilbene (PSB) and resveratrol (RES) on the gut microbiota brought by chronic jet‐lag in mice is investigated. Methods and Results Dietary supplement of RES and PSB (0.25%) are given to 16 week‐jetlagged mice to examine the effects on microbiota and physiological functions. Jetlag significantly induces weight gained that c… Show more

“…Gut microbiota analysis was performed according to the method described previously. 23 Isolating and purifying genomic DNA of the gut bacteria using an InnuPREP Stool DNA kit with slight modifications, the DNA samples were then sent to Biotools Co. Ltd for analysis of the fecal microbial composition via the 16S amplicon sequencing technique. PCR was used to amplify the 16S rRNA gene from each DNA sample, including 10 conserved regions (V3−V4) and 9 hypervariable regions (V1− V9).…”

“…Gut microbiota analysis was performed according to the method described previously. 23 Isolating and purifying genomic DNA of the gut bacteria using an InnuPREP The reads generated by sequencing on an Illumina HiSeq2500 platform (250 bp) formed effective tags that were clustered into ASVs (Amplicon Sequence Variants) at a 97% identity threshold, representing a taxonomic unit of bacterial species or genus. Weighted or unweighted uniFrac was used to quantify indexes such as PCA (principal components analysis) and PCoA (principal coordinates analysis), while LEfSe (linear discriminant analysis effect size) revealed the most identified differences among samples.…”
