Die Cardenolide von Parquetina nigrescens (AFZEL.) BULLOCK. Glykoside und Aglykone, 270. Mitteilung [1]

Abstract: Six crystalline (A, C, E, H, J and K) and six amorphous (α, γ, A1, D, D′, and G) but paperchromatographically homogenous cardenolides were isolated from the wood of Parquetina nigrescens (AFZEL.) BULLOCK (= Periploca nigrescens AFZEL.) after fermentation with glucosidases. The presence of twelve more cardenolides was observed in paperchromatograms. Three of the mentioned compounds could be identified with known substances: A = strophanthidin (already isolated by SCHENKER et al. [3] from similar material), H = … Show more

“…Strophanthidin.-A portion (27 mg) of fraction 12 was recrystallized from CHC13 to afford colorless crystals of strophanthidin, mp 138-l4l°/222-228°[lit. (25) 128-l42°/228-24l°], Ir and 'H-nmr spectra were identical to those of an authentic sample. Tic on silica gel with CHCl3-Me0H-H20 (70:26:4) EtOAc-cyclohexane (1:1), and MeOH-CHCl3 (15:75) and on alumina with MeOH-CHCl3 (15:75) and CHCl3-Me0H-H20 (70:26:4) gave Rf values identical with those of an authentic sample.…”

“…In 1954, the chemical investigation of P. nigrescens resulted in strophanthidin ( 28 ) and strophanthidol ( 29 ), with both containing an oxygenic substituent at C-19 [24]. Later, in 1957 and 1965, eight analogues ( 30 – 37 ) were isolated from the same species [25,26]. Compounds 35 – 37 are reduced by a pair of hydrogen atoms to form a double bond at C-16 and C-17 compared with 28 and 29 .…”

“…The leaves of P. nigrescens Afzel, (Periplocaceae) have been extensively used in traditional medicine as an antiinfective agent in East and West Africa (1). Previous work on P. nigrescens has resulted in the isolation of cardiac glycosides of the strophanthidin type from the stem and seeds (2,3), cymarin, monohydroxyolean-12-enecarboxylic acid, and a mixtrue of ci-and 3-amyrin from the roots (4), and calocin, genin, and plocigenin from the dried twigs (5).…”
