Abstract: Treatment with nonsteroidal anti-inflammatory drugs (NSAIDs) is associated with various side effects, including cardiovascular and hepatic disorders. Studies suggest that mitochondrial damage and oxidative stress are important mediators of toxicity, yet the underlying mechanisms are poorly understood. In this study, we identified that some NSAIDs, including diclofenac, inhibit autophagic flux in hepatocytes. Further detailed studies demonstrated that diclofenac induced a reactive oxygen species (ROS)-dependent… Show more

“…There are three types of autophagy – macroautophagy, microautophagy, and molecular chaperone-mediated autophagy – which transfer substances to lysosomes for degradation through different molecular pathways ( Li et al, 2021a ). According to the substance to be degraded in the autophagy vesicles, macroautophagy can be divided into mitochondrial autophagy (mitophagy), lipid autophagy, and endoplasmic reticulum autophagy ( Jung et al, 2020 ; Zhao et al, 2021b ).…”

“…Previous studies indicated that after long-term HFD feeding in mice, autophagosome biogenesis decreased markedly, resulting in impaired lipophagy (Yang, Li, Fu, Calay & Hotamisligil, 2010). However, recent studies found that a reduction in autophagic flux caused by the impairment of autophagosome-lysosome fusion led to inadequate lipophagy and intracellular lipid accumulation (Jung et al, 2020). In contrast, lipid accumulation in hepatocytes enhances autophagosome biogenesis.…”
