Development of a Specimen-Sparing Multichannel Bead Assay to Detect Antiparasite IgG4 for the Diagnosis of Schistosoma and Wuchereria Infections on the Coast of Kenya

Abstract: Abstract. To better delineate the impact of parasitic coinfection in coastal Kenya, we developed a novel specimensparing bead assay using multiplex flow immunoassay (MFI) technology to simultaneously measure serum or plasma immunoglobulin G4 (IgG4) against Brugia malayi antigen (BMA) and Schistosoma haematobium soluble worm antigen (SWAP). Properties of the bead assay were estimated by latent class analysis using data from S. haematobium egg counts/ filarial rapid diagnostic cards (RDTs), parasite-specific enz… Show more

“…To increase detection of low-intensity infections, collected serum and plasma and red blood cells were stored at −80°C and later tested by enzyme-linked immunosorbent assay for evidence of circulating filarial OG4C3 antigen (TropBioMed, Townsville, Australia) and anti-Schistosoma soluble worm antigen proteins immunoglobulin G4, diagnostic of current or recent infection. 22 Red blood cell pellets underwent DNA extraction for polymerase chain reaction speciesspecific detection of malaria parasites. 23 …”

“…Anti-Schistosoma IgG4 and anti-filarial IgG4, markers of active or very recent infection by these helminth parasites, were detected and quantified by specimen-sparing multiplex flow bead immunoassay as previously described. 22 Testing for anemia. Venous whole blood (4-5 mL) was collected from each subject by venipuncture.…”

“…6). As IgG4 in humans decreases dramatically after treatment with praziquantel, detection of IgG4 antibody responses against helminthic infections can help differentiate active infection from prior exposure [60]. Thus, further study of human schistosomiasis will be required with the aim of detecting specific IgG4 to SmTR antigens in sera and urine, thereby providing more insights into these currently hypothetical proteins.…”

“…IgG4 subtypes in human and IgG1 in mouse are involved in resistance and susceptibility to schistosomiasis [58,59] and thus are of great interest for the diagnosis of helminthic diseases [60].…”
