Development of a one-step analysis method for several amino acids using a microfluidic paper-based analytical device

Abstract: A one-step analysis method was developed for four types of amino acids using a microfluidic paper-based analytical device fabricated from chromatography filtration paper and laminate films. Aminoacyl-tRNA synthetase was used to detect each amino acid. The obtained laminated paper-based analytical device (LPAD) contained four enzymatic reaction areas. Colorimetric detection was performed based on the molybdenum blue reaction. A model method for the simple, easy, and simultaneous detection of several amino acid … Show more

“…More recently, Kugimiya et al [ 427 ] developed a laminated paper-based analytical device (LPAD), exploiting an aminoacyl-tRNA synthetase (aaRS) analysis system, to measure histidine, tryptophan, glycine, and lysine levels ( Figure 20 ). It included a sample spot connected to four enzymatic reaction areas, each containing a specific tRNA synthetase for one amino acid type and four detection areas.…”

“…These distinctive features make μPADs particularly appealing for POC testing in resource-limited and less-industrialized regions. Several reports have demonstrated the efficacy of μPADs in detecting a broad range of analytes, yielding promising results. …”

“…Paper-based analytical devices (PADs) are another good example of point-of-care diagnostics. They have evolved over the past decade due to properties such as low cost, biocompatibility, simple design and usage, and flexibility in pursuing affordable disposable testing units 4 6 . While sample analysis can be done in a few minutes, these devices have a relatively high limit of detection (LOD > µM), demanding amplification and cultivation time (usually several hours) for highly sensitive nucleic acid analysis.…”
