Determination of microphytobenthos PSII quantum efficiency and photosynthetic activity by means of variable chlorophyll fluorescence

Abstract: A pulse amplitude modulated fluorometer (PAM) was used to investigate photosynthetic activity of m~crophytobenthos on an Intertidal mudflat Spectral ~rradiance measurements Indicate that 7500 of the signal detectable by the PAM onginates In the upper 150 pm of the s e d~m e n t From the photosynthetic electron transport rate (ETR) measurements, it was concluded that the PAM could be used to observe changes in photosynthetic parameters during the day or the season Photoacclimation to lower irrad~ance was ind~ca… Show more

“…This result fits well with the ecology of these growth forms. EPL is not only more abundant in muddy cohesive sediments where light penetration is more restricted than in sandy sediments (Paterson and Hagerthey 2001;Cartaxana et al, 2011), but, more importantly, their (micro-) migratory behaviour allows positioning at the optimal irradiance in the vertical light gradient and rapid escape from periodic excess light (Kromkamp et al, 1998;Conn et al, 2004;Consalvey et al, 2004;Serô dio et al, 2006). This alleviates the need to invest in a strong physiological capacity to respond to light stress as previously proposed Cartaxana et al, 2011), although the right balance between motility and physiology still remains essential (van Leeuwe et al, 2009;Perkins et al, 2010b;Cartaxana et al, 2011;Serô dio et al, 2012).…”

“…Behavioural photoprotection involves motility, allowing cells to position themselves in light gradients and escape from prolonged exposure to excess light (Admiraal, 1984;Kromkamp et al, 1998;Consalvey et al, 2004;Serô dio et al, 2006). In addition, both motile and non-motile species use fast regulatory physiological processes for photoprotection (that is, 'physiological photoprotection'; Lavaud, 2007;Goss and Jakob, 2010;Depauw et al, 2012;Lepetit et al, 2012).…”

“…The larger pool size recorded in the Tagus than in the Eden estuary also corresponds with the higher average irradiance conditions in the Tagus (Table 1). Seasonal photoacclimation patterns of microphytobenthos have been reported previously (Blanchard & Cariou-Le Gall, 1994;Barranguet et al, 1998;Kromkamp et al, 1998), but the underlying physiological mechanisms have not been well described so far.…”

“…Accurate pigment data on microphytobenthic photoacclimation are still scarce and photosynthetic responses have been studied mainly by variable fluorescence analyses (e.g. Kromkamp et al, 1998;Perkins et al, 2001). Serodio et al (2005) were the first to measure the xanthophyll pigments and to link the presence of diatoxanthin to non-photochemical quenching signals.…”

“…Such a coefficient was estimated from the highest irradiance (690 µmol m -2 s -1 ) applied when obtaining the light response curves. The calculations followed Schreiber et al (1994) and Kromkamp et al (1998), and the terminology for chl fluorescence described by van Kooten and Snel (1990) was adopted. Curves of PI were generated on the basis of rETR and the respective parameters were calculated by the equation of Platt et al (1980): αETR, P max (rETR max ), I k , I c and photoinhibition parameter (βETR).…”
