Detection of DNA damage in fish Oreochromis mossambicus induced by co-exposure to phenanthrene and nitrite by ESI-MS/MS

Abstract: DNA adduct analysis in fish bile offers a promising approach to study the risk of potentiation of anthropogenic chemicals into genotoxic compounds in the presence of nitrite in the marine environment. We believe this is the first report on the formation of DNA-phenanthrene adducts on co-exposure of the fish to PAH and nitrite.

“…In fact, electrospray ionization tandem mass spectrometric (ESI-MS/MS) analysis was capable of revealing DNA adducts in different aquatic organisms (e.g. fish (Oreochromis mossambicus) and mussel (Perna perna) soft tissue) [185,186]. This MS/MS approach provided a rapid determination and discrimination of structurally different phenanthrene derived DNA adducts in fish bile samples [186].…”

“…Perhaps the sensitivity of different fish species to the toxic effects of nitrite is due to physiological characteristics of the regulatory mechanisms of ion homeostasis, determining levels of income and accumulation of exogenous nitrite in fish (Jensen 2007). Along with this, some authors suggest that nitrite, which have high reactivity in aqueous media may be involved in the formation of complex compounds with other xenobiotics-polycyclic aromatic hydrocarbons (Wahidulla and Rajamanickam 2010). Studies conducted by the authors quoted a source, demonstrating that the reaction products nitrosation polycyclic aromatic hydrocarbons can vary extremely high cytotoxicity.…”
