Detection of bat coronaviruses from Miniopterus fuliginosus in Japan

Abstract: Bats have great potential as reservoirs for emerging viruses such as severe acute respiratory syndrome-coronavirus. In this study, bat coronaviruses (BtCoVs) were detected by RT-PCR from intestinal and fecal specimens of Miniopterus fuliginosus breeding colonies in Wakayama Prefecture caves, where we previously identified bat betaherpesvirus 2. Two primer sets were used for the detection of BtCoV: one was for the RNA-dependent RNA polymerase (RdRp) region and the other was for the spike (S) protein region. Ele… Show more

“…These bats have a wide distribution ( 8 ), and increasing numbers of viruses, including coronaviruses and betaherpesviruses, are being isolated from them ( 9 , 10 ). Of the 6 bat species we sampled, only M. fuliginosus was positive for BtHV.…”

“…Saliva and nasal swabs were collected using UTM Virus Collection (360C, Copan Diagnosis, Brescia, Italy) depending on the breeding condition. Detection of MERS-CoV was performed by 2 MERS-CoV-specific assays-real-time RT-PCR (upE and ORF1 a probe sets) (10,11) and reverse transcription-loop-mediated isothermal amplification (RT-LAMP) (12) and a comprehensive assay using conventional RT-PCR using a primer set for pancoronaviruses (13). Viral RNAs were extracted using the QIAamp Viral RNA Mini Kit (Qiagen, Hilden, Germany) according to the manufacturer's instructions.…”

“…The supernatants were used for RNA isolation. The protocols for RT-PCR were previously described (10)(11)(12)(13). Five microliters of RNA were used for each amplification reaction.…”
