Detection and Quantification of the Capsular Polysaccharide of Burkholderia pseudomallei in Serum and Urine Samples from Melioidosis Patients

Abstract: Melioidosis is very challenging to diagnose. There is a clear need for a point-of-care assay for the detection of B. pseudomallei antigen directly from patient samples.

“…33,34 One study found that the detection of CPS in patient samples was possible, however, the sensitivity in serum and urine was low at 6.5% and 40.5% respectively. 15 A second study confirmed the low sensitivity of detecting CPS in serum samples and discussed that in resource-limited situations, the detection of CPS via LFI in serum may be useful. 23 Another study investigated LFI-based detection of CPS with various monoclonal antibodies and found that mAb 4C4 had a selective and sensitive affinity for CPS.…”

“…A calibrated dose response was developed using CPS-spiked serum or urine samples, and recovery was estimated from a linear graph. Biosensor properties such as dynamic linear range, accuracy, sensitivity, specificity, cross-reactivity, and interference were in each buffer and the results were compared to a CPS antigen-capture ELISA that has been previously described . The Bland-Altman test was performed to compare the results obtained using the READ sensor with ELISA.…”
