Detailed Analysis of the Human Mitochondrial Contact Site Complex Indicate a Hierarchy of Subunits

Abstract: Mitochondrial inner membrane folds into cristae, which significantly increase its surface and are important for mitochondrial function. The stability of cristae depends on the mitochondrial contact site (MICOS) complex. In human mitochondria, the inner membrane MICOS complex interacts with the outer membrane sorting and assembly machinery (SAM) complex, to form the mitochondrial intermembrane space bridging complex (MIB). We have created knockdown cell lines of most of the MICOS and MIB components and have use… Show more

“…4C). This may seem contradictory compared to previous observations in which the knockdown of Mic60/Mitofilin or Mic19/ CHCHD3 for 7 days induced depletion of the other MICOS subunits analyzed, whereas the knockdown of Mic25/CHCHD6, Mic27/ApoOL and Mic23/ApoO had no effect on other MICOS components (36). These conflicting results may be due to the fact that our cells were already stably depleted of hDISC1 once MICOS subunits expression was tested.…”

“…HeLa cells with inducible shRNA against Tob55/Sam50 or Tom40 49 (Figures 4a and b) were induced with DOX to obtain partial Tob55/Sam50 or Tom40 knockdown as they are essential proteins, 50 and used as target for parental YT-Indyexpressing GB. Three days of DOX induction was sufficient for the loss of Tob55/Sam50 without altering the expression of Tom40 and vice versa (Figures 4a and b).…”

“…Tumoral cells are characterized by the multistep acquisition of a series of specific hallmarks that ultimately lead to malignant transformation [44]. Among them the most represented are resistance to cell death, acquisition of uncontrolled proliferation and replicative immortality, metastatization and increased invasiveness, dysregulated cell metabolism and ROS production, mitochondrial quality control through mitophagy, together with tissue vascularization.…”
